P.Poeaim, A.The plant regeneration of teak (Tectona grandis L.) was reported. Result showed that the highest number of shoot induction was 55.49 cm., in length of shoots which tested at 0.25 mg·L <sup>-1</sup> BAP, and averaged 40.29 cm which tested at 0.25 mg·L <sup>-1</sup> mT after 4 weeks. The shoots were elongated using shoot tip explants of cryopreservation on teak by encapsulation-dehydration (EnDe), encapsulation-vitrification (EnVi) and vitrification (Vi) techniques. The shoot tips were used as explants, encapsulated apices were precultured on hormone-free MS medium containing 0.3 M sucrose for 2 days, dehydrated, and immersed in liquid nitrogen for 24 h after that thawed in thermal bath at 40 ºC and washed in unloading solution. Vi and EnVi protocols were used to treat with a loading solution, dehydrated by plant vitrification solution 2 (PVS2). EnDe protocol were encapsulated by desiccated air-dry in laminar air flow and dehydrated with silica gel. Results showed that Vi gave an excellent shoot tip explants for survival and regrowth. Vi were unable to survival. This experiment was preliminary searched to improve and develop the teak for cryopreservation. - Some of the metrics are blocked by yourconsent settings

P.Poeaim, A.Results described on shoot and root induction from node of medicinal plant Kadsura heteroclita. The sterilized nodes were used as explants and cultured on solid synthetic medium, Murashige and Skoog (MS) medium supplemented with single or combined Plant Growth Regulators (PGRs) of 6-benzylaminopurine (BAP), meta-Topolin (mT), Indole-3-butyric acid (IBA),1-Naphthaleneacetic acid (NAA) and Gibberellin (GA <inf>3</inf> ) were applied. The best result for shoot induction was shown on the medium supplemented with 0.5 mg/L of mT after 8 weeks. Transferred shoot to MS medium supplemented with combination PGRs of 0.5 mg/L of mT and 0.5, 1.0, 1.5 and 2.0 mg/L of IBA NAA or GA <inf>3</inf> after 16 weeks revealed that the explant were developed to multiple shoots which averaged number of shoot were 3 shoots/explant. The roots were not perfectly exhibited like sore on medium supplemented with 0.5 mg/L of mT and 1.5 mg/L of GA <inf>3</inf> . The high concentration of Auxin inhibited growth roots. The root induction was done by cutting the shoot and cultured on MS medium supplemented with combination PGRs of 0.5 mg/L of mT and 0.01, 0.02, 0.03, 0.04 and 0.05 mg/L of IBA after 4 weeks found that root length was approximately 2 mm on 0.5 mg/L of mT and 0.05 mg/L of IBA. - Some of the metrics are blocked by yourconsent settings
Item type:Publication, Efficiency of antioxidant and absorbent on browning and the optimal factors of plant regeneration from young seed of gluta usitata (217 Mae Ka) by Tissue Culture(2018-11-01) ;Rakrawee, R. ;Kittibanpacha, K. ;Chareonsap, P. P.Poeaim, A.Gluta usitata (Anacardiaceae) is a lacquer tree and create lacquer varnish can be utilized in many aspects especially art and culture. Recently, its harvest faced troublesome and yields of varnish tree are decreased. Consequently, the conservation of plant has reduced, and this plant has become extinct. The experiment was chosen to use plant tissue culture technique to preserve and increase this plant. The experiment was compared the effect of Woody Plant Medium (WPM) combined with different concentration of antioxidants were 2, 4, 6, 8 g/L Poly Vinyl Pyrrolidone (PVP), 20, 30, 40, 60 mg/L ascorbic acid or citric acid and absorbents of 1, 2, 3 g/L activated charcoal to reduce browning of phenolic compound in the explants. The results showed that 2 g/L activated charcoal gave the lowest of browning on seed and medium (about 0.9±0.2 points). The addition of 40 mg/L ascorbic acid was generated the highest of shoots (approximately 1.88±0.4 shoots/seed) after culture for 4 weeks. in vitro propagation of seed germination on WPM supplemented with 0.5, 1, 1.5, 2 and 3 mg/L 6-Benzyl amino purine (BAP), Thidiazuron (TDZ) and meta-Topolin (mT) 3% sucrose and 0.2% phytagel was efficiently resulted. Other result revealed that 1.5 mg/L BAP, 2 mg/L TDZ and 2 mg/L mT gave the highest seed germination at 1.6±0.2, 1.8±0.2 and 1.6±0.2 shoots/seed, respectively. The best of plant growth regulators for seed germination was 2 mg/L of TDZ after 6 weeks.
