    ZNO AND TIO2 nanoparticles stimulate callus induction and secondary metabolite production in indica RICE

    Metal nanoparticles are widely used in various fields of industry and their impact in the agricultural field is increasing. This research was carried out to investigate the effects of ZnO and TiO<inf>2</inf> nanoparticles on callus induction and plant metabolism in indica rice. Indica rice was assessed in plant tissue culture condition to estimate the influence of metal nanoparticles on morphological and physiological factors over 4 weeks. The seeds of Oryza sativa L. var. KDML105 were exposed to 1-3 mg L<sup>-1</sup> of 2,4-dichlorophenoxyacetic acid (2,4-D) in the callus induction medium. The induced-calli were cultured on the callus induction medium supplemented with 2 mg L<sup>-1</sup> of 2,4-D and showed a high increase in size and dry weight contents under the light condition when compared with the control and other treatments. Moreover, this research also investigated the potential impacts of ZnO and TiO<inf>2</inf> nanoparticles at 200, 400 and 800 mg L<sup>-1</sup> on morphological and metabolic responses of the induced-calli cultured on the callus induction medium supplemented with 2,4-D and each type of metal nanoparticles. At 4 weeks after treatment, the induced-calli were analyzed for plant growth and assays on aspects of their metabolism were undertaken. The induced-calli exposed to 200 mg L<sup>-1</sup> of ZnO or TiO<inf>2</inf> nanoparticles were found to have significant increase of biomass (size and dry weight of callus), total phenolic compounds and total flavonoid contents in KDML105 variety. The results demonstrated that utilization of ZnO and TiO<inf>2</inf> nanoparticles at appropriate concentration could positively induce the production of beneficial secondary metabolites in the induced-calli of indica rice and are worthy of further study for applications in other plant species.

    Improvement of Plant Regeneration Frequency from Carbon Sources in Aromatic Rice (Oryza sativa L.)

    The objective of this research is to investigate carbon sources for optimum callus induction and plant regeneration frequencies. The effect of carbon sources on callus induction and plant regeneration from mature seeds of aromatic rice (Oryza sativa L.) cultivar Khao Dawk Mali 105 (KDML105) was studied in this experiment. For callus induction, the different types of carbon sources along with 30 g L<sup>−1</sup> of sucrose or maltose were evaluated. NSI and NMI medium were used for callus induction in three replicates (sucrose and maltose as carbon sources, respectively). The calli derived from NMI medium showed the higher percentage of callus induction, fresh weight, dry weights, callus size, green spots and regeneration frequency than calli derived from NSI medium. For plant regeneration, the 3-week-old calli were transferred to the regeneration medium which was different in carbon sources (NSR, NMR, NSMR0.5, NMMR0.5, NSMR1, NMMR1, NSMR1.5, NMMR1.5, NSMR2, NMMR2, NSMR2.5 and NMMR2.5 medium) and tested to investigate the effect of carbon sources in three replicates. The percentage of regeneration (60.71%) and the ratio of the number of seedlings to the number of regenerated calli (3.02) obtained from NMR medium showed the highest values. The percentage of callus induction and morphological performance (weight and size) were correlated with plant regeneration frequency. The results showed that using maltose as a carbon source gave a better result than using sucrose as a carbon source for callus induction and plant regeneration in aromatic rice cultivar KDML105. The regenerated plants were successfully rooted and well grown in greenhouse conditions.

    Application of activated charcoal and nanocarbon to callus induction and plant regeneration in aromatic rice (Oryza sativa L.)

    The investigations of nanotechnology with the application on agricultural products also have been few reported, especially the plant regeneration. The effects of activated charcoal and nanocarbon on the callus induction and plant regeneration of aromatic rice were studied. Activated charcoal was added into the callus induction and regeneration medium. The presence of activated charcoal in the callus induction medium (100-500 mg L<sup>−1</sup>), activated charcoal significantly reduced the percentage of the callus induction and biomass accumulation (fresh weight, dry weight and size). Whereas, the regeneration medium supplemented with 100 mg L<sup>−1</sup> of activated charcoal showed the highest percentage of plant regeneration (61.90%) and the ratio of the number of seedlings to the number of regenerated calli (RSR; 3.06) that derived from the callus induction medium (without activated charcoal). Moreover, the induced calli derived from the callus induction medium supplemented with nanocarbon at 5 mg L<sup>−1</sup> showed the highest percentage of callus induction (94.70%), the percentage of green spots (95.83%), the percentage of plant regeneration (60.42%) and the RSR (3.12) when transferred the calli into the regeneration medium (without nanocarbon). After that, nanocarbon was also added into the regeneration medium. The percentage of green spots (96.08%), the percentage of plant regeneration (62.75%) and the RSR (3.16) obtained from the regeneration medium supplemented with 20 mg L<sup>−1</sup> of nanocarbon showed the highest values. This experiment showed that the optimum concentration of activated charcoal and nanocarbon had potential to enhance the callus induction and plant regeneration frequencies in tissue culture medium of aromatic rice.
