    Prediction of the germination rate and antioxidant properties of VD20 Rice by utilizing Artificial neural network-coupled response surface methodology and product characterization

    The current research aims to predict and optimize process conditions to produce germinated VD20 with a high rate of germination and antioxidant properties. Box-Behnken design (BBD) was used to build models with three factors [soaking time (ST: 4–6 h), germination time (GT: 18–22 h), and germination temperature (33–37 °C)] and three replications. The data set from the BBD experiment was used to create an artificial neural network (ANN) model that estimated the change in responses by variable factors. The ANN model was extremely accurate, with an overall correlation coefficient (R) of 0.9997 and showed the best fit with actual and predicted data. The germination conditions were further optimized using multi-objective RSM, which revealed that the optimal conditions were ST of 5.34 h, GT of 20.78 h, and germination temperature of 35.6 °C. The experimental validation revealed a high level of agreement between the results of the BBD models forecasted and the actual experimental values. The ANN-coupled BBD methodology is a promising hybrid method for modeling, forecasting, and optimizing the impact of process conditions on the quality of germinated grain. In addition, the raw and germinated VD20 rice were analyzed for their techno-functional properties, estimated glycemic index (eGI), and FTIR. Lower peak viscosity, values of breakdown and setback, and phytic acid were found after rice was germinated. The result revealed that high antioxidant content and activity, which were confirmed by the FTIR pattern, and low digestion behaviors (eGI = 64.23) were the attributes of the germinated product. Furthermore, the results of pasting, thermal, swelling power, and solubility showed the wide range of further application of this material, which should receive more consideration in future research.

    Characterization of Nipa Palm’s (Nypa fruticans Wurmb.) Sap and Syrup as Functional Food Ingredients

    Nipa palm (Nypa fruticans Wurmb.) sap and its syrup locally produced in southern, Thailand, were characterized. Sugar was the predominant dry matter of both sap and syrup (p < 0.05). Sucrose, glucose and fructose were detected in both samples. In both samples, potassium was the most abundant macroelement (p < 0.05), followed by sodium, phosphorus, magnesium and calcium, respectively. Iron, copper, zinc, manganese and iodine were also found. Syrup had higher total soluble solid, lower a<inf>w</inf> and pH, darker color with higher intermediate browning product (A<inf>285</inf>) and browning intensity (A<inf>420</inf>) than sap. Syrup had lower total phenolic and flavonoid contents than sap (p < 0.05). Vanillic acid was predominant in sap, whereas vanillic acid, rutin and trans-ferullic acid were identified in syrup. DPPH<sup>·</sup>-scavenging and ferrous ion-chelating abilities of sap were higher than syrup, whereas the ferric reducing antioxidant power of the syrup was higher (p < 0.05). Sap and syrup were able to inhibit α-glucosidase with the IC<inf>50</inf> of 15.79 and 222.51 mg/mL, respectively. Similar FTIR spectra with the different intensities of sap and syrup were noticeable suggesting the identical compositions of both samples with different contents. The peaks at 3380 and 2924 cm<sup>−1</sup> suggested the presence of carbohydrates and the bands at 1500–800 cm<sup>−1</sup> represented the zone of the three major sugar including sucrose, fructose and glucose. Our result suggests that nipa palm sap and syrup can be used as potential sources of functional food ingredients.

    Characterization of acid soluble collagen from the skin of snakeskin gourami (Trichogaster Pectoralis)

    The present study was aimed to isolate and characterized acid soluble collagen (ASC) from the skin of snakeskin gourami (Trichogaster pectoralis). ASC from gourami skin had a yield of 9.43% and 34.65%, based on wet and dry weight basis, respectively. The purity of ASC was superior with a distinct absorption peak at wavelength (WL) of 230.7 nm. Based on the electrophoretic pattern, gourami skin ASC was classified as type I collagen, as it comprised α1 and α2 as major components and higher molecular weight (MW) components γ, β were distinctly observed. ASC exhibited high Tmax value of 33.43°C, which could correspond to its imino acids content of 188 residues/1000 residues. Fourier transform infrared (FTIR) spectrum and circular dichroism (CD) revealed that ASC extracted from gourami skin had greater structural integrity in its triple-helical form. Solubility of ASC was high at the pH range of 2-4 in which zeta potential exhibited highly positive charge. The highest solubility of ASC in the presence of NaCl was observed at 2% (w/v). Therefore, with all the characteristic features, ASC from snakeskin gourami skin can be a valueadded product in the fish processing industry.

    Physicochemical characteristics of glucosamine from blue swimming crab (Portunus pelagicus) shell prepared by acid hydrolysis

    The aim of this research was to characterize glucosamine hydrochloride (GluHCl) from the shell of blue swimming crab (Portunus pelagicus). The crab shell was finely milled and processed to chitin prior to HCl hydrolysis using 30 % HCl for 30 min at 100 ºC for glucosamine production. The resultant glucosamine was recovered by crystallization using 95 % ethanol and was dried in a hot air oven. The color of the glucosamine crystals, expressed as L*, a*, and b*, was 83.01, 5.03, and-3.38, respectively. Crab shell glucosamine had high purity, which could be strongly stained by ninhydrin and presented at the same Rf of standard D-glucosamine using thin layer chromatography. Furthermore, prepared glucosamine exhibited similar Fourier transform infrared (FTIR) spectrum as standard D-glucosamine. Glucosamine from blue swimming crab shell had high purity as determined by HPLC and contained 808.15 mg D-glucosamine/g sample. The maximal transition temperature (T<inf>max</inf>) and the total enthalpy (ΔH) of prepared glucosamine were 194 ºC and 754.42 J/g, respectively. As a consequence, with the presented method, the resultant glucosamine was characterized to be D-glucosamine. Therefore, blue swimming crab shell, a byproduct from crab meat processing, has high potential as a raw material to produce glucosamine for food and nutraceutical applications.

    Characterization, functionality and antioxidant activity of water-soluble proteins extracted from bombyx mori linn.

    The yield of water-soluble protein extracted from silkworm pupae, Bombyx mori Linn. (WSPB) was 3.96% by wet weight basis. The major amino acids found in WSPB were glutamic acid, which was the most abundant, followed by histidine, phenylalanine and glycine in that order. The electrophoretic study revealed that proteins with MW of 37, 64 and 75 kDa were the major protein components in WSPB. Based on FTIR analysis, WSPB remained its structural integrity. The surface hydrophobicity, free and total sulfhydryl group contents were 3.52, 22.17 µmol/g and 23.08 µmol/g, respectively. WSPB was highly solubilized in the pH range of 5-11. WSPB exhibited poor emulsifying properties and foaming capacity but the foam stability was comparable to bovine serum albumin (BSA). WSPB had high antioxidant potential, based on DPPH <sup>•</sup> , ABTS <sup>•+</sup> and FRAP assay. Therefore, protein from silkworm pupae is a potential source of antioxidant and can be served as an ingredient in processed foods to enhance its desired functionality and nutritional value.

    Purification and characterization of a novel plantaricin, KL-1Y, from Lactobacillus plantarum KL-1

    Three bacteriocins from Lactobacillus plantarum KL-1 were successfully purified using ammonium sulfate precipitation, cation-exchange chromatography and reverse-phase HPLC. The bacteriocin peptides KL-1X, -1Y and -1Z had molecular masses of 3053.82, 3498.16 and 3533.16 Da, respectively. All three peptides were stable at pH 2–12 and 25 °C and at high temperatures of 80 and 100 °C for 30 min and 121 °C for 15 min. However, they differed in their susceptibility to proteolytic enzymes and their inhibition spectra. KL-1Y showed broad inhibitory activities against Gram-positive and Gram-negative bacteria, including Salmonella enterica serovar Enteritidis DMST 17368, Pseudomonas aeruginosa ATCC 15442, P. aeruginosa ATCC 9027, Escherichia coli O157:H7 and E. coli ATCC 8739. KL-1X and -1Z inhibited only Gram-positive bacteria. KL-1X, KL-1Y and KL-1Z exhibited synergistic activity. The successful amino acid sequencing of KL-1Y had a hydrophobicity of approximately 30 % and no cysteine residues suggested its novelty, and it was designated “plantaricin KL-1Y”. Plantaricin KL-1Y exhibited bactericidal activity against Bacillus cereus JCM 2152<sup>T</sup>. Compared to nisin, KL-1Y displayed broad inhibitory activities of 200, 800, 1600, 800, 400 and 400 AU/mL against the growth of Bacillus coagulans JCM 2257<sup>T</sup>, B. cereus JCM 2152<sup>T</sup>, Listeria innocua ATCC 33090<sup>T</sup>, Staphylococcus aureus TISTR 118, E. coli O157:H7 and E. coli ATCC 8739, respectively, whereas nisin had similar activities against only B. coagulans JCM 2257<sup>T</sup> and B. cereus JCM 2152<sup>T</sup>. Therefore, the novel plantaricin KL-1Y is a promising antimicrobial substance for food safety uses in the future.

    Facile synthesis and characterization of tenorite nanoparticles from gas-atomized Cu powder

    CuO powder is widely used because of its high activity and selectivity in oxidation/reduction reactions. CuO nanoparticles are interesting because of their high ratio of surface area to mass, which is expected to enhance the catalytic performance of a material. Because of such importance, CuO nanoparticles were prepared by two different facile routes. The first involved the reaction of pure Cu powder with nitric acid to form Cu(NO<inf>3</inf>)<inf>2</inf> intermediate. In the second route, glacial acetic acid was employed to react with pure Cu powder to form Cu(CH<inf>3</inf>COO)<inf>2</inf> intermediate. After purification, both intermediates were reacted with NaOH via a solid-state route to form CuO powders. The intermediates and end products were characterized by Fourier transform infrared spectroscopy, scanning electron microscopy and X-ray diffraction technique. Experimental results indicated that synthesis of the CuO nanoparticles via the Cu(NO<inf>3</inf>)<inf>2</inf> intermediate yielded slightly higher product and fine particle sizes with a larger aspect ratio. In contrast, the Cu(CH<inf>3</inf>COO)<inf>2</inf> intermediate route yielded lower product yield and fine particle sizes with a smaller aspect ratio. However, due to the extremely fine particle size it was observed that nanoparticle agglomeration could not be avoided particularly in the CuO nanopowders produced via the Cu(CH<inf>3</inf>COO)<inf>2</inf> intermediate route.

    Facile, alternative synthesis of spherical-like ca(H2PO4)2•H2O nanoparticle by aqueous-methanol media

    Spherical-like calcium dihydrogenphosphate monohydrate (Ca(H<inf>2</inf>PO<inf>4</inf>)<inf>2</inf>•H<inf>2</inf>O) nanostructure was successfully prepared by the mixing of calcium carbonate and phosphoric in aqueous-methanol media at ambient temperature for 30 min. Three thermal decomposition step and higher stability at over 800°C of the prepared sample are different from the earlier works. Spherical-like Ca(H<inf>2</inf>PO<inf>4</inf>)<inf>2</inf>•H<inf>2</inf>O nanostructure with diameter < 100 nm confirmed by SEM may be important for potential applications. This method of synthesis by aqueous-methanol media is a fast and simple method and it is expected to be applicable for the synthesis of other nanocrystalline calcium phosphates. © (2013) Trans Tech Publications, Switzerland.

    Non-isothermal decomposition kinetics of NiFe2O4 nanoparticles synthesized using egg white solution route

    The thermal decomposition kinetics of nickel ferrite (NiFe <inf>2</inf>O<inf>4</inf>) precursor prepared using egg white solution route in dynamical air atmosphere was studied by means of TG with different heating rates. The activation energy (E <inf>α</inf>) values of one reaction process were estimated using the methods of Flynn-Wall-Ozawa (FWO) and Kissinger-Akahira-Sunose (KAS), which were found to be consistent. The dependent activation energies on extent of conversions of the decomposition reaction indicate "multi-step" processes. XRD, SEM and FTIR showed that the synthesized NiFe<inf>2</inf>O<inf>4</inf> precursor after calcination at 773 K has a pure spinel phase, having particle sizes of ~54 ± 29 nm. © 2009 Akadémiai Kiadó, Budapest, Hungary.
