Item type:Publication, Plant regeneration of Bauhinia purpurea by tissue culture technique(2024-05-01) ;Saparam, W. ;Poeaim, A. ;Pongtongkam, P. ;Chareonsap, P. P.Results showed that the seed of Bauhinia purpurea germination was archived 100% from half-strength MS medium supplemented with 3.0 mg/L Gibberellic acids (GA3) and gave 71.137 mm for the highest seedling. The optimal condition of nodal explants sterilization was E5, which gave 92% of survival explants. For the shoot induction method, the results showed that 24.270 mm was the highest proliferation of shoots achieved from MS medium supplemented with 1.0 mg/L 6-benzylaminopurine (BAP). The highest number of roots was 2.867 roots, and the highest root length was 33.453 mm, obtained at half-strength MS medium complemented with 0.25 mg/L Indole-3-acetic acid (IAA). P. ;Poeaim, A.The experimental findings demonstrated that the highest proliferation frequency (100%), along with the maximum number of shoots per explant (3.33±0.52 shoots) and an average shoot length of 30.04±1.70 mm, were achieved in the Murashige and Skoog (MS) medium supplemented with 2.0 mg/l of 6-benzylaminopurine (BAP). Subsequently, upon reaching a height of 3 to 4 cm, the in vitro micro shoots were transferred to a ¼ MS medium enriched with 0.75 mg/l of indole-3-acetic acid (IAA). This particular regimen resulted in the highest number of roots per shootlet (3.50±0.58) with an average root length of 34.19±0.79 mm. The acclimatization phase of the plantlets to the soil was successful, demonstrating a commendable survival rate of 80%. - Some of the metrics are blocked by yourconsent settings
Item type:Publication, In vitro effect of gamma irradiation and plant growth regulators (PGRs) for induction and development of Stylosanthes hamata cv. Verano(2019-01-01) ;Ngoenngam, L. ;Pongtongkam, P. ;Arananant, J.; Poeam, A.Pasture legumes was developed by tissue culture technique using gamma irradiation. Seeds of Stylosanthes hamata cv. Verano were received 7 doses of radiation (5, 10, 15, 20, 25, 30 and 45 Krad) and control (without irradiation) cultured on MS medium. Free Plant Growth Regulators (PGRs) and 30 g/L sucrose treatment found that 30 Krad decreased the percentage of shoot and root length of 31.7% and 20.51% respectively in 30 days. While seeds without irradiation were 100% germination. The radiation doses was reduced the growth rate by half in 7 days GR <inf>50</inf> (3 <inf>0)</inf> at 39.82 Krad. The lethal dose (LD <inf>50</inf> ) of Stylosanthes hamata cv. Verano was 26.14 Krad and the higher irradiation level increased death rate. Seeds on MS medium with 3 mg/L of meta-Topolin (mT) resulted at 60 days after irradiation which higher than 50 % in all treatments. P. ;Poeaim, A.It was found that the sterilization procedure was highly effective, resulting in a 100% survival rate of Xylia xylocarpa (Roxb.). After sterilization, the seeds were cultured on a half-strength Murashige and Skoog (MS) medium without adding plant growth regulators (PGRs). A maximum germination percentage of 100% was achieved after 2 weeks of culture. 1.5 cm-sized node segments from both seeds and mother trees were used for shoot induction. These node segments were treated with the same sterilization conditions as the seeds but with the addition of 0.1% HgCl2. The node segments achieved a 90% survival rate despite the additional chemicals. The node segments from seeds were then cultured on a full-strength MS medium supplemented with 0.50 mg/l of BAP. The results showed a maximum number of shoots, reaching 4.10±1.20 per explant and a mean shoot length of 3.87±0.30 cm. The part of node segments derived from mother trees showed 3.50±1.27 shoots per explant, accompanied by a mean shoot length of 3.18±0.89 cm when using BAP concentration of 0.25 mg/l after 12 weeks of cultivation. The rooting medium contained half-strength MS medium supplemented with 0.75 mg/l of IBA. After 8 weeks of being cultured in the rooting medium, the shoots exhibited a maximum rooting percentage of about 80%. The technique provides a valuable method for rapidly propagating and conserving this plant species in a controlled and efficient manner. - Some of the metrics are blocked by yourconsent settings
Item type:Publication, In vitro effect of plant growth regulators (PGRs) for callus induction and plant regeneration from suspension of Hamata (Stylosanthes hamata cv. Verano)(2018-12-01) ;Ngoenngam, L. ;Pongtongkam, P. ;Arananant, J.; Poeaim, A.The plant regeneration from seed of Hamata (Stylosanthes hamata cv. Verano) was found. Seeds were induced on MS medium (Murashige and Skoog, 1962) with different concentration of cytokinin were used 0.5, 1 and 3 mg/L 6-benzyladenine (BAP) meta-Topolin (mT) or Thidizuron (TDZ), respectively. The result showed that the highest percentage of seed induction was 65% and average shoots 8.66 shoots per seed on MS medium with 3 mg/L mT for 12 weeks cultivation. For node of hamata were study on callus and shoot induction on MS medium with 0.5 mg/L IBA combined with 0.5, 1 and 3 mg/L TDZ. The highest percentage of callus and shoot induction was 100% gave average areas of callus at 11192.84 mm <sup>3</sup> and the number of shoots per callus was 13.50 shoots on MS medium with 0.5 mg/L IBA combined 3 mg/L TDZ for 8 weeks cultivation Cell suspension culture of callus that developed from node of hamata. The friable callus was transferred to liquid MS medium with 0.5 mg/L IBA combined with 3 mg/L TDZ for 4 weeks cultivation After that transferred to MS medium with 3 mg/L mT for shoots regeneration. The percentage of shoots generation from callus was shown on 90% and the number of shoots per callus was 14.00 shoots for 12 weeks cultivation. Transferred shoot to root medium containing of MS medium with Indole-3-butyric acid (IBA) Naphthaleneacetic acid (NAA) or Indole-3-acetic acid (IAA) at 0.1, 0.3 and 0.5 mg/L. The result showed that the highest percentage of root induction was 100% and gave averaged roots at 11.00 <sup>+</sup> 1.87 roots per shoot on MS medium with 0.3 mg/L NAA for 12 weeks cultivation. - Some of the metrics are blocked by yourconsent settings
Item type:Publication, Effects of growth regulators on in vitro propagation of Sophora tomentosa L. (Necklace pod)(2024-05-01) ;Rungprateeppaiboon, N. ;Chareonsap, P. P. ;Pongtongkam, P. ;Poeaim, A.Seeds of Sophora tomentosa were sterilized and germinated on MS medium with BAP (1 mg/L), which was observed to be 75% germination rate with the highest amount of multiple shoots (3 shoots per seed) and an average length of 29.56 mm. The germination rate was reduced to 33.33% by 1% NaCl. Multiple shoot induction was worked on MS medium with 2.0 mg/L of TDZ that induced the highest shoot bud formation rate (46.67%) with the highest amount of shoot bud (10.71 buds per culture), while 1.0 mg/L GA3 showed the highest average length of 34.13 mm. For root induction, 20% rooting was formed in 1/2 MS medium with a combination of 0.5 mg/L IAA and BAP with 0.1% AC, with 3 roots per culture and an average root length of 15.36 mm. P.The minimum contamination percentage (25%) was recorded in the experiment using 0.2% mercuric chloride (HgCl2) for 20 min, which was observed to be a 50% germination rate after 2 weeks of culture. The maximum germination rate (68%) was obtained on Murashige and Skoog (MS) solid medium supplemented with 1.0 mg/L 6-benzylaminopurine (BAP), giving a shoot length of 48.00 mm. and root length of 31.40 mm. The optimal condition of nodal segment sterilization was recorded in the experiment using 0.1% HgCl2 for 10 min, which gave 80% survival explants. For shoot induction, it was worked on MS solid medium with 0.5 mg/L 6-benzylaminopurine (BAP) that induced the highest average length of 11.31 mm. The highest root formation frequency (50%) and root length (3.0 mm) were achieved on an MS medium containing 2.0 mg/L indole-3-butyric acid (IBA). The plantlets were acclimatized and established in the soil.
