    Effect of different aging methods and periods on physical and chemical beef qualities from Longissimus thoracis

    The results showed that the aging method and aging period significantly (p<0.05) affected aging loss, trimming loss, and cooking loss. Aging loss and trimming loss of DA and DB beef were higher than those of WV and WS beef (p<0.05). However, cooking loss in WV and WS beef was higher than DA and DB beef. No significant differences (p>0.05) were observed among aging methods in pH, colour (L*, a*, and b*), Warner-Bratzler shear force (WBSF), lipid oxidation (TBARS). Prolonged aging time (up to 4 weeks) resulted in more tender meat, while lipid oxidation increased (p<0.05). Additionally, beef colour changed significantly with increasing aging time (p<0.05). There were not different in percentage of fat, dry matter, ash, or moisture among aging method. However, DA and AB beef showed higher percentage of protein than WV and WS beef (p<0.05). These findings can be applied to develop for beef industry, particularly small-scale beef retailers and restaurants businesses, could be enhance value of premium grade beef in Thailand.

    Potential antioxidant and lipid peroxidation inhibition of Phyllanthus acidus leaf extract in minced pork

    Objective: This study investigated the effect of extraction solvents on antioxidant bio-active compounds as well as potential antioxidant and lipid peroxidation inhibition of Phyllanthus acidus (P. acidus) leaf extract in minced pork. Methods: The effect of various solvent systems of water, 25%, 50%, 75% (v/v) ethanol in water and absolute ethanol on the extraction crude yield, total phenolic content, total flavonoid content and in vitro antioxidant activities of P. acidus leaves was determined. In addition, antioxidant activities of the addition of crude extract from P. aciuds leaves at 2.5 and 5 g/kg in minced pork on 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging, 2,2'-Azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS) radical cation decolorization, reducing power and inhibition of lipid peroxidation (thiobarbituric acid reactive substances; TBARS) were determined. Moreover, sensory evaluation of the samples was undertaken by using a 7-point hedonic scale. Results: The results showed that the highest crude yield (2.8 g/100 g dry weight) was obtained from water which also had the highest recovery yield for total phenolic content, total flavonoid content and the strongest antioxidant activity. The addition of crude water extract from P. acidus leaves was more effective in retarding lipid peroxidation and higher antioxidant activity than control and butylated hydroxytoluene in minced pork. In particular, the samples containing P. acidus extract had no significant effect on the sensory scores of overall appearance, color, odor, texture, flavor, and overall acceptability compared to the control. Conclusion: Water solvent was an optimally appropriate solvent for P. acidus leaf extraction because of its ability to yield the highest amount of bio-active compounds and in vitro antioxidant property. Particularly, P. acidus crude water extract also strongly expressed the capacity to retard lipid oxidation, radical scavenging, radical cation decolorization and reducing power in minced pork. The results of this study indicated that P. acidus leaf extract could be used as natural antioxidant in the pork industry.

    Enhancing health-oriented Chashu pork: Effects of transglutaminase and omega-3 enriched pork belly with improved sliceability and fatty acid profile

    Results from the first experiment indicated that powdered transglutaminase (TG) at 1% w/w resulted in the highest slicing yield, hardness, and gumminess of the fat portion compared with the control and dissolved TG treatments (P<0.05). Sensory evaluation also showed significantly higher scores for binding, taste, texture, and overall acceptability than the other treatments (P<0.05). The second experiment further investigated Chashu pork produced from omega-3 enriched pork in comparison with pork obtained from animals fed a standard diet (control). Analysis of composition showed that omega-3 pork and its Chashu contained lower fat but higher protein and moisture contents as well as significantly greater omega-3 fatty acids compared with control pork (P<0.05). In addition, these samples demonstrated a higher polyunsaturated fatty acids/saturated fatty acids (PUFA/SFA) ratio (P<0.05), indicating improved lipid nutritional quality. Notably, Chashu from omega-3 pork had approximately 3.8-fold higher omega-3 fatty acids than control Chashu and achieved an omega-6/omega-3 ratio below 4.0, compared with about 19.3 in the control group. Sensory evaluation revealed no significant differences in all tested attributes between treatments (P>0.05). These findings demonstrated that the use of powdered TG improved the sliceability and sensory quality of Chashu pork, while omega-3 pork provided additional nutritional benefits by improving the PUFA/SFA ratio and enhancing omega-3 fatty acids. This approach not only helps to reduce product loss caused by separation during slicing, but also enhances the value of pork belly with adding health benefits.

    In vitro study of antioxidant and antimicrobial activities of soybean tempeh and split gill fungus (Schizophyllum commune) as plant-based diets

    Plant-based diets that reduce or eliminate animal product intake are known to ameliorate the incidence and clinical course of several illnesses. Soybean tempeh and split gill edible fungus (Schizophyllum commune) have long been consumed as plant-based diet choices. Total phenolic content (TPC), antioxidant and antimicrobial activities of soybean tempeh and S. commune extracts were investigated using water for extraction. The aqueous extract of fresh S. commune had the highest TPC at 1191.81 mg GAE/100g. Three methods were evaluated antioxidant activity as the diphenyl picrylhydrazyl radical scavenging test (DPPH), radical cation decolorization assay (ABTS) and reducing power. The IC<inf>50</inf> values of fresh S. commune using DPPH and ABTS+ assays were 0.2812% and 0.5761%, respectively whereas the reducing power assay gave EC<inf>50</inf> value at 0.3606%. Crude extracts of fresh tempeh and fresh S. commune exhibited antimicrobial activity against Staphylococcus aureus TISTR 746 at concentrations of 25 and 50 mg/ml, respectively. Two times the minimum inhibitory concentration (MIC) of fresh tempeh and fresh S. commune effectively reduced viable cells of S. aureus TISTR 746 of 5 log CFU/ml at 12 h. Both fresh tempeh and S. commune extracts showed promise as natural antioxidants for utilization as plant-based food.

    Siamese neem tree as a natural preservative: Chemical profile, antioxidant properties, and antibacterial efficacy against foodborne pathogens and spoilage bacteria

    The objective of the present study was to investigate the effect of various ethanol in water solvent systems (0, 25, 50, 75 and 100 % v/v) on the extraction crude yield, total phenolic content, total flavonoid content, and chemical profile of Azadirachta indica (Siamese neem tree) leaf extracts. In addition, the antioxidant and antimicrobial properties were investigated using in vitro assays. Crude yield was highest for the water extract (5.78 g/100 g DW) and decreased with increased ethanol. Total phenolic and flavonoid contents were highest in the 75 % ethanol extract (26.95 mg GAE/g crude and 23.73 mg QE/g crude, respectively). GC-MS analysis of the 75 % ethanol extract revealed 33 bioactive compounds, accounting for 96.33 % of the total area. Significant components included benzene, 1,4-bis(phenylmethyl)- (15.05 %), and naphthalene, 1,6-dimethyl- (10.98 %); also present were sterols, fatty acids, hydrocarbons, aromatic, and heterocyclic compounds, indicating a complex chemical profile. In antioxidant assays, the 75 % ethanol extract revealed the highest DPPH radical scavenging activity (IC<inf>50</inf>: 143.03 mg/L), reducing power (IC<inf>50</inf>: 378.69 mg/L), and lipid peroxidation (IC<inf>50</inf>: 526.25 mg/L). However, the absolute ethanol extract exhibited both the highest metal chelating activity (IC<inf>50</inf>: 382.60 mg/L) and the strongest antimicrobial activity, inhibiting all 11 tested foodborne pathogenic and spoilage bacteria. The lowest minimum inhibitory concentration (MIC) of 25 mg/mL was observed for Staphylococcus aureus, Aeromonas hydrophila, Pseudomonas fluorescens, Latilactobacillus sakei, Streptococcus sp., and Lactococcus cremoris, with inhibition zone diameters of 27.21, 28.79, 28.19, 17.60, 15.09 and 11.20 mm, respectively. Similarly, the lowest minimum bactericidal concentration (MBC) of 25 mg/mL was obtained for S. aureus and L. sakei, indicating potent antibacterial effects against these species. Future studies should include direct comparative analyses of neem extracts and synthetic preservatives to evaluate further their relative efficacy and applicability in real-world food systems.

    Characterization of antimicrobial substance from Lactobacillus salivarius KL-D4 and its application as biopreservative for creamy filling

    Lactobacillus salivarius KL-D4 isolated from duck intestine produced bacteriocin which was stable at high temperature and a wide pH range of 3–10. Its cell free supernatant at pH 5.5 exhibited wide inhibitory spectrum against both G+ and G− bacteria. The highest bacteriocin production was obtained in MRS broth supplemented with 0.5 % (w/v) CaCO<inf>3</inf> at 6 h by gentle shaking. PCR walking using specific primers at the conserved region of class-II bacteriocin resulted in 4 known genes of kld1, kld2, kld3 and kld4 with 100 % similarity to genes encoding for salivaricin α, β, induction peptide and histidine protein kinase of Lb. salivarius GJ-24 which did not previously report for bacteriocin characterization, while showing 94, 93, 59 and 62 % to other salivaricin gene cluster, respectively. The high activities of 25,600 AU/ml indicated a strong induction peptide expressed by kld3 which has low similarity to previous inducer reported. Based on operon analysis, only kld1, kld3 and kld4 could be expressed and subsequently elucidated that only salivaricin α like bacteriocin was produced and secreted out of the cells. Using protein purification, only a single peptide band obtained showed that this strain produced one bacteriocin which could be salivaricin α namely salivaricin KLD showing about 4.3 kDa on SDS-PAGE. Partial purification by 20 % ammonium sulfate precipitation of the product was tested on the artificial contamination of creamy filling by Bacillus cereus, Enterococcus faecalis, Pseudomonas stutzeri, Staphylococcus sp. and Stenotrophomonas sp. resulting the growth inhibitory efficiency of 4.45–66.9, 11.5–100, 100, 0–28.1 and 5–100 % respectively. Therefore, salivaricin KLD can be a tentative biopreservative for food industry in the future.

    Effects of humectant and roasting on physicochemical and sensory properties of jerky made from spent hen meat

    This study was carried out to develop jerky product utilizing spent hen meat. Manipulation of water activity (aw) of semi-dried meat with the addition of humectants (glycerol and sorbitol at the concentration of 0, 10, and 15%) and roasting process were applied to enhance quality of jerky product. After spent hen meat was ground, formulated, reformed and then dried in convection oven at 85oC for 2 h followed by 60oC for 1 h, all jerky samples showed values of aw ≤0.85 with total aerobic bacteria, yeast and mold and Staphylococcus aureus counts were reduced to undetectable levels. Moreover, the results showed that the 15% glycerol added jerky showed superior quality, indicated by the lowest aw, low protein aggregation as evaluated by sorption isotherm and the lowest shear value as compared to sorbitol added and control samples (p<0.05). Regardless effect of humectant, roasting could improve the quality of the jerky via enhanced sensory attributes by increasing color, appearance, flavor and overall acceptability scores (p<0.05). Therefore, spent hen meat could be used as a potential raw material for jerky in which soft product texture obtained by the addition of glycerol and intense flavor resulted from roasting process after drying, leading to sensory acceptance.

    In vitro antioxidant activities and volatile compounds from karanda (Carissa carandas L.) fruit wine

    Carissa carandas L. or karanda (in Thai called namdaeng or manaao ho) is widely used as a medicinal plant. Karanda juice was fermented for 22 days to produce alcohol using Saccharomyces cerevisiae TISTR5918. Total soluble solids or TSS of karanda juice (approximately 1.0°Brix) was adjusted to 24°Brix and samples were collected every 2 days for analysis of total soluble solids (TSS), pH and alcohol content. Yield of alcohol was measured at 12.50±0.35% in the final fermentation. TSS dropped gradually from 24°Brix to 9.17±0.58°Brix, whereas pH increased slightly from 2.47±0.06 to 2.80±0.00 (Day 0-22). Volatile compounds in karanda wine (KW) were examined and analyzed using gas chromatography-mass spectrometry (GC-MS). Volatile compounds detected in KW included 3-methyl-1-butanol, butanoic acid, butyrolactone, phenylethyl alcohol, butanedioic acid, benzene ethanol and 2-propenyl ester as a common flavor and aroma in wine. Heavy metals (Pb, As, Cu and Cd) determined by atomic absorption spectroscopy (AAS) were present in lower than their maximum allowed concentrations. Methanol was detected at only 0.005% in KW analyzed by gas chromatography equipped with a flame ionization detector (GC-FID). Antioxidant activities were determined by three methods comprising diphenyl picrylhydrazyl radical scavenging assay (DPPH), radical cation decolorization assay (ABTS) and reducing power. The IC <inf>50</inf> values of DPPH and ABTS were 0.84±0.03% and 3.28±0.03%, respectively, and the EC <inf>50</inf> of reducing power was 11.03±0.11%. Total phenolic content, measured according to the Folin-Ciocalteu procedure, was determined as 746.64±3.10 mg GAE/100 ml sample.

    Application of tempeh and split gill mushroom extracts in herbal fresh sausage: Evolution of antioxidant and antimicrobial activities

    Tempeh, recognized as a high-protein meat alternative, is gaining popularity across Asian countries, while split gill mushroom (Schizophyllum commune), rich in essential nutrients, are increasingly consumed in Southeast Asia. The antioxidant activities of all treatments were significantly higher than the control (p<0.05). Furthermore, formulations containing split gill mushroom (R3, R4, and R5) exhibited lower initial total plate count (TPC) values compared to the other formulations. Sensory evaluation was then conducted on sausages containing tempeh, split gill mushrooms, and their combinations in ratios of 1:1, 1:3, and 3:1, yielding overall acceptance scores of 3.93, 3.30, 4.27, 5.33, and 3.37, respectively. The combination with a 3:1 tempeh-to-mushroom ratio demonstrated the highest consumer acceptance. Therefore, the study on the application of tempeh and split gill mushroom extracts in herbal fresh sausage is provided valuable information for developing plant-based products and functional foods in the future.

    In vitro screening of potential probiotic lactic acid bacteria isolated from intestinal contents and gills of Nile tilapia

    The potential probiotic lactic acid bacteria (LAB) were isolated from intestinal contents and gills of Nile tilapia (Oreochromis niloticus). Those properties included antagonistic activity against pathogenic bacteria, tolerance to pH, NaCl, fresh bile from Nile tilapia. Moreover, drug resistance profiles were examined. A total of forty isolates were collected. LAB isolated from the intestinal contents showed inhibitory activity against Bacillus coagulans TISTR1447, Pseudomonas fluorescens TISTR358 and Salmonella Typhimurium TISTR292, whereas those from the gills revealed only antagonistic activity against Bacillus coagulans TISTR1447, S. Typhimurium TISTR292 and Escherichia coli TISTR780. Isolates LI10 and LG5 were selected for further evaluation of probiotic properties. Both isolates could grow in pH 4-10, NaCl concentration up to 10% and fresh bile concentration up to 10%. These isolates were resistant to cefovecin, marbofloxacin, clindamycin and trimethoprim/sulfamethoxazole, meanwhile they were susceptible to benzylpenicillin, amoxicillin/clavulanic acid, vancomycin, tetracycline, nitrofurantoin and chloramphenicol. In addition, both isolates were identified by Matrix-assisted lazer desorption/ionization-time of flight mass spectrometry (MALDI-TOF MS) as Enterococcus feacalis. Therefore, these two isolates could potentially be used as probiotics.
