Derivative spectrophotometric determination of acetaminophen by the formation of [Fe(II)-(2,2′-bipyridyl)3]2+

Abstract
A sensitive, stability-indicating and precise second-derivative spectrophotometric method assay for acetaminophen in paracetamol tablets has been developed based upon the reduction of Fe(III) to Fe(II) by the acetaminophen in the presence with 2,2′-bipyridyl forming an orange-red chelate complex, [Fe(II)-(2,2′-bipyridyl)3]2+, λmax at 522 nm. The linear concentration range is 0-12 μg mL-1. The detection limit is 0.176 μg mL-1 and the recovery was 100.5%. The method has been successfully applied to the assay of commercial paracetamol tablets and results compared with those obtained by the USP XXII official method.
