    ZNO AND TIO2 nanoparticles stimulate callus induction and secondary metabolite production in indica RICE

    Metal nanoparticles are widely used in various fields of industry and their impact in the agricultural field is increasing. This research was carried out to investigate the effects of ZnO and TiO<inf>2</inf> nanoparticles on callus induction and plant metabolism in indica rice. Indica rice was assessed in plant tissue culture condition to estimate the influence of metal nanoparticles on morphological and physiological factors over 4 weeks. The seeds of Oryza sativa L. var. KDML105 were exposed to 1-3 mg L<sup>-1</sup> of 2,4-dichlorophenoxyacetic acid (2,4-D) in the callus induction medium. The induced-calli were cultured on the callus induction medium supplemented with 2 mg L<sup>-1</sup> of 2,4-D and showed a high increase in size and dry weight contents under the light condition when compared with the control and other treatments. Moreover, this research also investigated the potential impacts of ZnO and TiO<inf>2</inf> nanoparticles at 200, 400 and 800 mg L<sup>-1</sup> on morphological and metabolic responses of the induced-calli cultured on the callus induction medium supplemented with 2,4-D and each type of metal nanoparticles. At 4 weeks after treatment, the induced-calli were analyzed for plant growth and assays on aspects of their metabolism were undertaken. The induced-calli exposed to 200 mg L<sup>-1</sup> of ZnO or TiO<inf>2</inf> nanoparticles were found to have significant increase of biomass (size and dry weight of callus), total phenolic compounds and total flavonoid contents in KDML105 variety. The results demonstrated that utilization of ZnO and TiO<inf>2</inf> nanoparticles at appropriate concentration could positively induce the production of beneficial secondary metabolites in the induced-calli of indica rice and are worthy of further study for applications in other plant species.

    Efficient plant regeneration systems are useful for indica rice using micropropagation and/or genetic transformation. Nanomaterials provide applications and opportunities in various fields of life science and agriculture technology. In this research, indica rice cultivars ‘Suphanburi1’ and ‘Suphanburi90’ (Oryza sativa L. ssp. indica) were used to investigate. Seeds were examined to evaluate callus induction and plant regeneration frequency. Culture of sterilized-seeds on callus induction medium supplemented with 50 mg L<sup>-1</sup> of TiO<inf>2</inf> nanoparticles induced a high frequency of callus induction and content of biomass accumulation. Embryogenic calli were transferred to plant regeneration medium supplemented with 10-50 mg L<sup>-1</sup> TiO<inf>2</inf> nanoparticles. The highest frequency of plant regeneration and the ratio of the number of seedlings to the number of regenerated calli were produced on plant regeneration medium supplemented with 40 mg L<sup>-1</sup> of TiO<inf>2</inf> nanoparticles. The regenerated shoots induced roots on NB medium without plant growth regulators and the plantlets were established in soil. The results may be used for nanomaterial applications of rice and/or other plants for its further improvement.

    Modifications of morphological and physiological characteristics of pigmented-rice seedlings by application of titanium dioxide nanoparticles

    Nanomaterials or metal nanoparticles are considered as the agricultural applications with significant response-promoting influence. This research was focused on the influence of titanium dioxide nanoparticles (TiO<inf>2</inf> NPs) on morphological and physiological processes in pigmented-rice plants at seedling stage. A significant influence of TiO<inf>2</inf> NPs on the plant growth, photosynthetic pigment contents and antioxidant enzyme activities in rice seedlings was investigated. The results were also shown that pigmented-rice seedlings growing in TiO<inf>2</inf> NPs application significantly differed in the morphological and physiological responses when compared with the control plants. The results presented the plant height and dry weight was significantly affected by TiO<inf>2</inf> NPs. The results observed an increase in chlorophyll A, chlorophyll B, carotenoid contents and catalase, peroxidase enzyme activities in plant seedlings of TiO<inf>2</inf> NPs-treated plants which positive-corresponded with plant growth performances was found. TiO<inf>2</inf> NPs-treated seedlings that a high level of TiO<inf>2</inf> NPs application exhibited an increase in the plant growth, photosynthetic pigment contents and antioxidant enzyme activities. The amelioration effect of 800 mg L<sup>-1</sup> of TiO<inf>2</inf> NPs application was shown the highest performances in morphological and physiological responses when compared with the other treatments. Investigation of the results suggests that the morphological and physiological responses due to changes in the plant growth, photosynthetic pigment content and antioxidant enzyme activity parameters may derive from the interaction between TiO<inf>2</inf> NPs and plant cells of pigmented-rice seedlings, which are known to play a role in plant development and productivity in the further study.

    Assessment of antioxidant enzymes in response to exogenous titanium dioxide (TiO2) nanoparticles in Chainat 1 rice cultivar

    Antioxidant enzymes are important to prevent the destruction of photosynthetic pigments in plant cells. There have been reported that ZnO nanoparticles can promote activities of antioxidant enzymes in rice, but should not use in high activities because it can cause toxicity. Therefore, the objective of this study was to investigate the effect of titanium dioxide (TiO<inf>2</inf>) nanoparticles on the activities of antioxidant enzymes in Chainat 1 rice cultivar. Rice plants were treated with TiO<inf>2</inf> nanoparticles using different concentrations i.e., 0, 100, 200, 400, 600 and 800 mg.L<sup>-1</sup> for every 7 days-interval. Photosynthetic pigment contents (chlorophyll A, chlorophyll B and carotenoid) and activities of antioxidant enzymes (catalase and peroxidase) were recorded at 37, 44 and 51 days after planting. The results showed that the treatment of TiO<inf>2</inf> nanoparticles was not effect on the quantities of chlorophyll A, chlorophyll B and carotenoids, but found that the activities of peroxidase and catalase enzymes were significant affected at all growth stages. Especially, at a concentration of 200 mg.L<sup>-1</sup> TiO<inf>2</inf> found that rice plants contained the highest activities of peroxidase enzyme and significant difference from control treatment. However, the treatments of higher dose than 200 mg.L<sup>-1</sup> TiO<inf>2</inf> would have a chance to meet toxicity in rice plants as the activities of antioxidant enzymes were decreased.

    Application of nanomaterials in plant regeneration of rice (Oryza sativa L.)

    In recent years, the application of nanomaterials has been investigated by various studies, especially in their biological and agricultural features. This research aimed to evaluate the influence of zinc oxide (ZnO) and titanium dioxide (TiO<inf>2</inf>) nanoparticles on the quality of the plant regeneration in rice (Oryza sativa L. cv. RD49). Mature seed explants were used for callus induction and plant regeneration cultured on the medium supplemented with different concentrations of ZnO or TiO<inf>2</inf> nanoparticles. The high percentage of the callus induction was achieved on the medium supplemented with 5 mg.L<sup>-1</sup> ZnO nanoparticles or 20 mg.L<sup>-1</sup> TiO<inf>2</inf> nanoparticles when compared with control treatment (without nanoparticles). Various concentrations of TiO<inf>2</inf> nanoparticles were also found to increase the percentage of the plant regeneration more than ZnO nanoparticles. The highest percentage of plant regeneration was observed on the medium containing the concentration of 20 mg.L<sup>-1</sup> TiO<inf>2</inf> nanoparticles. The in vitro rooting of regenerated shoots was hardened and successfully transplanted to the field after 30 days of transplantation. The results suggested that ZnO and TiO<inf>2</inf> nanomaterials may have potential applications in the in vitro plant regeneration.
