Item type:Publication, Alpha-amylase inhibitory activity of collagen hydrolysate from Asian bullfrog skin and its application in dark chocolate(2024-01-01) ;Indriani, Sylvia ;Benjakul, Soottawat ;Sitanggang, Azis Boing ;Karnjanapratum, SupatraNalinanon, SitthipongThe present study aimed to investigate the in-vitro antidiabetic activity of collagen hydrolysate (CH) from Asian bullfrog skin and its application as a bioactive compound in dark chocolate bars. Papain (3% [w/w]) was used to obtain CH with a simultaneous conjunction of ultrasound. CH possessed α-amylase inhibitory activity with an IC<inf>50</inf> value of 3.60 mg/mL, while results from inhibition reaction kinetics revealed that CH inhibited α-amylase in a non-competitive mode. Different concentrations (0.5, 1.0, and 2.0% [w/w]) of CH were then fortified into dark chocolate bars, and the physicochemical characteristics, breaking strength, and rheological behaviour of the resulting chocolate were investigated and compared with a control (without CH addition). A higher level of CH significantly enhanced antidiabetic activity against α-amylase (p ≤ 0.05) in chocolate bar. Chocolate bar with 2% CH complied with the nutritional claim requirement for ‘source of protein’ with lower fat content and energy value, while sensory evaluation revealed good organoleptic properties. - Some of the metrics are blocked by yourconsent settings
Item type:Publication, Structural characteristic and molecular docking simulation of fish protein-derived peptides: Recent updates on antioxidant, anti-hypertensive and anti-diabetic peptides(2023-03-30) ;Prakash Nirmal, Nilesh ;Singh Rajput, Mithun ;Bhojraj Rathod, Nikheel ;Mudgil, PritiPati, SiddharthaOver the decade, fish protein-derived peptides (FPDP) have been evaluated for various biological activities including their mechanism of action through structure–activity relationship (SAR) and molecular simulation. SAR studies are known to provide the basic structural information of the active site which can be used for designing synthetic bioactive peptides for application in therapeutics and medicinal purposes. In light of the above discussion, this review discusses the mechanism of action and SAR of the FPDP with a focus on three widely studied bioactive properties including antioxidant, antihypertensive and anti-diabetic activities. The emphasis is given to the recently purified and identified FPDP from various seafood resources. A brief discussion has been made on their structural characteristics and mechanism of action towards antioxidant, angiotensin-I converting enzyme (ACE) inhibition, and dipeptidyl peptidase-IV (DPP-IV) inhibitory activities. The present study investigated the in vitro inhibition of α-amylase, α-glucosidase, and lipase, and antioxidant activities (DPPH, metal chelating, and FRAP) of Allium cepa L., Apium graveolens L., Coriandrum sativum L., and Petroselinum crispum (Mill.) Fuss using solvent 80% ethanol, absolute ethanol, acetone, methanol, n-hexane, chloroform, and hot water. The total phenolic content (TPC) and the FTIR and GC-MS characterization spectra were compared with the activities. The TPC of 80% ethanol extract in A. graveolens L. was the highest among the extracts, with 23.78 mg GAE/g extract. The 80% ethanol extract exhibited the most potent antioxidant activity on DPPH radical scavenging (A. graveolens, 125.57 mg AEAC/g extract), metal chelating (C. sativum, 92.85 mg EECC/g extract), and FRAP activities (C. sativum, 46.98 mg AEAC/g extract). The 80% ethanol extract showed the highest anti-α-amylase (P. crispum, 30.61 mmol ACE/g extract) and anti-α-glucosidase (A. cepa, 595.28 mmol ACE/g extract) activities. In comparison, the absolute ethanol extract of C. sativum showed the highest anti-lipase activity (42.10% inhibition). According to the FTIR spectra, the 80% ethanol extract of the four green leafy vegetables is predicted to have several active compounds. GC-MS identified the compounds responsible for the activity. Studies on the recovery potential of active compounds as oral agents to treat diabetes mellitus with certain solvents are very useful in producing oral agents and functional foods to prevent diabetes mellitus. - Some of the metrics are blocked by yourconsent settings
Item type:Publication, Evaluation of antidiabetic, antioxidant and other phytochemical properties of Thai fruits, vegetables and some local food plants(2019-01-01) ;Nanasombat, Suree ;Yansodthee, KanokwanJongjaited, IladaAntidiabetic, antioxidant, anti-acetylcholinesterase and prebiotic activities, total phenolics and flavonoids of 33 crude ethanolic extracts of Thai local fruits, vegetables and some local food plants were determined. Mangosteen (Garcinia mangostana) fruit peel and Indian gooseberry (Phyllanthus emblica) fruit extracts had highest antioxidant activity. Bamboo grass (Tiliacora triandra) leaf extract had strongest α-amylase inhibitory activity (78.28 % inhibition), while mulberry (Morus alba) fruit extract had strongest α-glucosidase inhibitory activity (59.63 % inhibition). Star cactus (Aloe vera) leaf pulp extract had strongest anti-acetylcholinesterase activity (31.55 % inhibition). Indian gooseberry fruit and mangosteen fruit peel extracts had highest content of total phenolics and flavonoids, respectively. The extract with highest indigestible polysaccharide content was the extract of mangosteen fruit peels (188.62 mg/g extract), while those with relatively high indigestible polysaccharides were the extracts of pineapple fruits (Ananas comosus), lotus seeds (Nelumbo nucifera), black rice grains (Oryza sativa) and pisang mas banana fruits (Musa sapientum). Based on these properties, 5 plant extracts were selected to study for their prebiotic effect on growth and fermentation of Lactobacillus acidophilus, Lactobacillus bulgaricus and Streptococcus thermophilus in yogurt at 42 °C. Addition of lotus seed extract resulted in highest proliferation of these bacteria with 2.24 logCFU/g increase of total lactic acid bacterial counts in yogurt after 24 h fermentation, while addition of black rice grain, pisang mas banana fruit and pineapple fruit extracts caused good growth of these bacteria with 2.12-2.19 logCFU/g increase of total counts.
