    The multiple shoots induction of Stylosanthes guianensis CIAT 184 was cultured on MS medium supplemented with 3 mg/l TDZ. The percentage of shoot induction was 100%, the average number of shoots was 36.80 per seed, and the average height of shoots was 0.76 cm. The shoots were transferred to MS medium for rooting, supplemented with 0.3 mg/l IAA. The percentage of root induction was 40%, and the average number of roots was 6.50 per shoot. Physical gamma rays induced the mutation. Seeds were irradiated with 0, 5, 10, 15, 20, 25, 30 and 50 Krad. The results showed that S. guianensis CIAT 184 irradiated at 44.38 Krad gave 50 % (GR50(30)) and 50% LD50(30) of 38.06 Krad. The drought tolerance of seeds supplemented with 0, 5, 10, 15 and 20% of PEG-6000. The GR50 was 17.73 % at 2 weeks, and the LD50 was 14.86 % and 11.83 % for 4 and 12 weeks, respectively.

    The results of solid Gamborg's B5 medium at 2 mg/L mT found the highest average number of shoots at 1.6 shoots per explant; and the number of leaves at 3.7 leaves per explant. The concentration of 1.5 mg/L of mT showed the highest average length of shoots which was 9.82 mm, while the 0.5 mg/L mT revealed the most significant number of roots which were 0.9 roots per explant with the longest root of 3.75 mm. The liquid Gamborg's B5 medium showed the highest average number of shoots at 1.5 mg/L BAP, the same as at 1.5 mg/L mT. The medium without plant growth regulator and 1 mg/L mT was the highest average length of shoots of 13.98 mm. The highest number of roots per explant was 0.8, similar to 2 mg/L mT, but the different maximum length of roots of 3.04 mm. A hundred percent of protocorms induced shoots found in all media. The plantlets were survived after transplanting into pots and covered with plastic bags in four weeks.

    Improvement of Plant Regeneration Frequency from Carbon Sources in Aromatic Rice (Oryza sativa L.)
    (2018-09-01) ;
    Sutjaritvorakul, Thanawat
    The objective of this research is to investigate carbon sources for optimum callus induction and plant regeneration frequencies. The effect of carbon sources on callus induction and plant regeneration from mature seeds of aromatic rice (Oryza sativa L.) cultivar Khao Dawk Mali 105 (KDML105) was studied in this experiment. For callus induction, the different types of carbon sources along with 30 g L−1 of sucrose or maltose were evaluated. NSI and NMI medium were used for callus induction in three replicates (sucrose and maltose as carbon sources, respectively). The calli derived from NMI medium showed the higher percentage of callus induction, fresh weight, dry weights, callus size, green spots and regeneration frequency than calli derived from NSI medium. For plant regeneration, the 3-week-old calli were transferred to the regeneration medium which was different in carbon sources (NSR, NMR, NSMR0.5, NMMR0.5, NSMR1, NMMR1, NSMR1.5, NMMR1.5, NSMR2, NMMR2, NSMR2.5 and NMMR2.5 medium) and tested to investigate the effect of carbon sources in three replicates. The percentage of regeneration (60.71%) and the ratio of the number of seedlings to the number of regenerated calli (3.02) obtained from NMR medium showed the highest values. The percentage of callus induction and morphological performance (weight and size) were correlated with plant regeneration frequency. The results showed that using maltose as a carbon source gave a better result than using sucrose as a carbon source for callus induction and plant regeneration in aromatic rice cultivar KDML105. The regenerated plants were successfully rooted and well grown in greenhouse conditions.

    Synergistic effects of BAP and kinetin media additives on regeneration of vetiver grass (Vetiveria zizanioides L. Nash)
    (2016-05-01) ;
    Khunchuay, Chonnikarn
    Regeneration efficiency was enhanced by adding auxin and cytokinin at different concentrations to the media. It was found that the two highest frequencies-90 and 85%-of callus induction occured when the buds were cultured in an MS medium supplemented with 2 mgL-1 a-naphthalene acetic acid (NAA) plus 1 mgL-1 N6-benzylaminopurine (BAP) (CIM2) and in the same medium supplemented with 2 mgL-1 NAA plus 1 mgL-1 kinetin (CIM5), respectively. When cultured further in a plant regeneration medium supplemented with 1 mgL-1 NAA and 2 mgL-1 BAP (PRM1), these calli regenerated with high efficiencies, 80% for calli grown in CIM2 and 85% for calli grown in CIM5. The low BAP concentration in CIM1 (2 mgL-1 NAA and 0.5 mgL-1 BAP) resulted in a low regeneration efficiency and fewer shoots per callus even though the PRM3 regeneration medium had a high concentration of cytokinins (2 mgL-1 BAP and 2 mgL-1 kinetin) . However, the calli cultured in CIM2 without kinetin but with a suitable level of BAP still regenerated with high efficiencies in PRM2 and PRM3 that contained 1 and 2 mgL-1 kinetin, respectively. Similarly, when calli cultured in CIM5 without BAP but with kinetin were regenerated in PRM1 with BAP alone, they also regenerated with a high regeneration efficiency. These results showed that culturing a specific tissue in a medium with suitable concentrations of NAA and BAP or kinetin resulted in a large number of multiple shoots, and BAP and kinetin worked synergistically to produce a high plant regeneration efficiency.
