The growing interest in bacteriocin-producing lactic acid bacteria from traditional fermented foods has highlighted the value of strains that combine antimicrobial activity, probiotic functionality, and genome-supported safety. This study presents an integrated functional and probiogenomic characterization of Pediococcus pentosaceus NR4-4 isolated from Thai fermented mustard greens. The strain was found to produce a heat-stable class IIa bacteriocin with pronounced inhibitory activity against Listeria monocytogenes . In addition, NR4-4 exhibited strong tolerance to simulated gastrointestinal conditions and demonstrated effective adhesion to Caco-2 intestinal epithelial cells. In vitro safety evaluations confirmed that the strain was non-hemolytic and did not exhibit cytotoxic effects toward AGS or Caco-2 cell lines. Whole-genome sequencing verified species identity and revealed the presence of pediocin-related gene clusters, carbohydrate-active enzymes associated with fructooligosaccharide utilization, and a favorable genome-based safety profile. Notably, no virulence factors, biogenic amine biosynthesis pathways, plasmids, or transferable antimicrobial resistance determinants were detected. The preferential metabolism of fructooligosaccharides further suggests compatibility with synbiotic formulations. Overall, these findings indicate that P. pentosaceus NR4-4 is a genomically supported, safe, and multifunctional probiotic candidate with potential applications in food biopreservation and functional food development.

Biodegradable packaging coated with antimicrobial compounds has emerged as a new type of food packaging with potential to enhance food safety, extend shelf life and minimize plastic pollution. We have developed a polylactic acid (PLA) composite pineapple leaf fibre (PALF) biopolymer to fulfil the high demand of biodegradable materials for a potential use in food packaging. Our formulation with 10% pineapple fibre PLA/PALF composites exhibited enhanced melting temperatures and heat resistance, with a 44% reduction in the tensile strength and 2.7-fold reduction on the strain at the break when compared with neat PLA. To impart antimicrobial activity, the PLA/PALF was coated with crude supernatants isolated from Pediococcus pentosaceus PP04 containing 1.3% w/v of nisin. This material coated with the crude extract was shown to inhibit the growth of Bacillus cereus ATCC 11778, Salmonella enterica subsp. enterica serovar Typhimurium ATCC 13311, and Escherichia coli ATCC 8739 in culture conditions, as well as minimizing the number of the tested pathogens on coated material surfaces.

Lactic acid bacteria (LAB) from raw and fermented pork samples were screened for their inhibitory activity by an agar spot test in order to obtain a LAB strain with suitable property to be used as meat starter cultures. Among the 174 isolates, 73 were positive to inhibit at least one of the seven indicator bacteria, which were further characterized. The most suitable isolate was isolate P0805, identified as Pediococcus pentosaceus. This bacterium was catalase- and nitrate reductase-positive and amino acid decarboxylase-negative; moreover, it produced inhibitory substances against Salmonella Typhimurium with the activity of the partially purified inhibitory substances of 409,600 AU/mL. To further characterize the catalase-producing ability of P. pentosaceus P0805, the effect of hematin on its catalase activity in Sausage Model Broth (SMB) was evaluated, and it enhanced catalase production. The catalase activity was found in both SMB with and without hematin. It was concluded that catalase produced by this bacterium was heme-independent catalase.
