Upland rice vinegar vapor inhibits spore germination, hyphal growth and aflatoxin formation in Aspergillus flavus on maize grains
The efficacy of vapor-phase (VP) upland rice vinegar (URV) was investigated as a bio-fumigant for maize, to reduce consumer health risks associated with spore and toxin formation by Aspergillus flavus. Complete reduction of mycelial growth occurred with in vitro VP exposure to URV (containing 0.0017 mmol/L acetic acid) or with VP exposure to pure acetic acid (PAA) (containing 0.0023 mmol/L acetic acid). No significant differences were observed between the two materials after 90 min exposures. Using gas chromatography-mass spectrometry (GC-MS), URV vapor was shown to contain volatiles having antifungal activities. These are identified as isoamylalcohol, 1-butanol, 3-methyl-, acetate and β-phenylethyl acetate. It is suggested these volatiles increase the antifungal effectiveness of URV. Exposure to VP-URV (containing 0.0043 mmol/L AA) for 5 h completely eliminated viable spores of A. flavus on maize seeds (23% moisture content) previously inoculated with 4.43 ± 0.28 log spores/g). At the same time, aflatoxin production decreased, as VP-URV exposure increased. Hence, VP-URV is shown to be an effective control agent for A. flavus mycelial growth and aflatoxin formation on maize, so effectively reducing the potential for consumer health risks due to this widespread fungus.

Microalgae show promise as a biomass and bioproduct for applications in various industries. The cultivation of microalgae plays a crucial role in optimizing biomass yield and bioproduct accumulation. The provision of inorganic carbon substrates substantially enhances microalgal growth and lipid biosynthesis, resulting in marked increases in the production of biofuels and other bioproducts. This study examined biomass and lipid accumulation in Messastrum gracile IFRPD 1061 under inorganic stress conditions, previously unreported. M. gracile IFRPD 1061 was subjected to varying conditions of inorganic carbon substrates, 1–3 g·L−1 sodium carbonate and bicarbonate concentration, to enhance biomass and lipid accumulation. Optimal productivity levels were observed with sodium bicarbonate addition of 3 g·L−1 and 1 g·L−1 for biomass and lipids, resulting in productivities of 392.64 and 53.57 mg·L−1·d−1, respectively. Results underlined the effectiveness of sodium carbonate and bicarbonate as inorganic carbon sources for stimulating microalgal growth and enhancing the production of high-value products. The extraction of lipids from freeze-dried biomass of M. gracile IFRPD 1061 demonstrated optimal yield using methanol/hexane solvents compared with the control experiments. Lipid extraction yields using methanol/hexane were 42.18% and 46.81% from oven-dried and freeze-dried biomass, respectively. Lipids extracted from oven-dried M. gracile IFRPD 1061 using methanol/hexane/chloroform solvents indicated the potential of methanol/hexane as a solvent for lipid extraction from dry microalgal biomass using an ultrasonic-assisted technique. This study contributes valuable insights into maximizing biofuel and bioproduct production from microalgae, highlighting A. gracilis as a promising candidate for industrial applications.

Rhodotorula yeast has potential in terms of biotechnology for the production of various compounds for the environmental, industrial, and medical sectors. It is well known to generate carotenoid pigments that are beneficially distinguished. This investigation aimed to optimize the growth of Rhodotorula paludigena CM33 and its production of β-carotene. Response Surface Methodology was employed to optimize temperature, ammonium sulfate, and ferric chloride concentrations in a 5L-bioreactor batch culture. The optimal conditions of 30 °C, 6.2 g/L of ammonium sulfate, and 75.0 mg/L of ferric chloride provided biomass of 17.71 g/L, a specific growth rate of 0.22 h−1, and β-carotene concentration of 40.29 mg/L. Upon scaling up the cultivation process, the 22L-bioreactor displayed similar growth patterns to those of the 5L-bioreactor. However, the 22L-bioreactor exhibited higher product yields in a shorter time period. The maximum biomass production of 18.96 ± 1.31 g/L and the maximum β-carotene production of 42.81 ± 0.61 mg/L were achieved at 28 h. This scaling-up strategy resulted in improved production efficiency. Additionally, the nutritional value of the cells was evaluated, suggesting their potential as animal feed supplements. R. paludigena CM33 demonstrated a β-carotene single-cell protein content, making it a valuable source for aquaculture and livestock feed. These findings highlight the potential of this yeast strain for large-scale production of β-carotene and single-cell protein, offering nutritional benefits for animal feed.

This study investigates the feasibility of using soy milk as a substrate for cultivating Lactobacillus casei and its subsequent conversion into paraprobiotics. The research focuses on optimizing the fermentation process from shake flasks to bioreactor scale and assessing the antioxidant properties and proximate compositions of the resulting paraprobiotic soy milk beverage. The study highlights a dual-step large-scale fermentation process comprising an anaerobic fermentation step followed by a thermal inactivation step using the in situ temperature control system of the bioreactor. Results indicated that soy milk enriched with 10 g/L glucose and fermented at 37 °C for 24 h on flask scale provided optimal conditions for L. casei growth, achieving a bacterial count increase of 8.80 ± 0.11 log CFU/mL and a titratable acidity of 0.89 ± 0.03 %. These optimal conditions were employed for bioreactor scale fermentation, attaining a bacterial count of 10.02 ± 0.06 log CFU/mL and a titratable acidity of 1.07 ± 0.04 % at 24 h. The thermal inactivation step at 90 °C for 30 min successfully converted the probiotic soy milk into a paraprobiotic form, maintaining its antioxidant activity at 62.12 ± 2.58 % radical scavenging activity. The final paraprobiotic product met the Food and Drug Administration (FDA) specifications for titratable acidity, presenting a viable alternative for soy-based functional beverages.

The instability of the protein expression in Pichia pastoris strains has been an issue for various peptide productions. Some modifications to the traditional fermentation process could potentially solve the problem. Here, we consider a four-stage fermentation process to express the CAP2 (cell-penetrating antimicrobial peptide 2) candidate in P. pastoris KM71H, a slow methanol utilization strain. During the fermentation process, CAP2 productivity is limited (6.15 ± 0.21 mg/L·h) by the low overall methanol consumption (approximately 645 g), which is mainly the result of the slow methanol utilization of the P. pastoris KM71H. To overcome this limitation, we increased the cell concentration two-fold prior to the induction stage. A fed-batch process with exponential and dissolved oxygen tension (DOT) stat feeding strategies was deployed to control the glycerol feed, resulting in an increase in cell concentration and enhancement of the volumetric methanol consumption rate. The improved fermentation process increased the overall methanol consumption (approximately 1070 g) and the CAP2 productivity (13.59 ± 0.24 mg/L·h) by 1.66 and 2.21 times, respectively. In addition, the CAP3 (cell-penetrating antimicrobial peptide 3) candidate could also be produced using this improved fermentation process at a high yield of 3.96 ± 0.02 g/L without any further optimization. Note that there was no oxygen limitation during the improved fermentation process operating at high cell density. This could be due to the controlled substrate addition via the DOT stat system.

Fermented plant-based products were gathered from various regions in Thailand and categorized into 10 types of traditional commercial vegetables. Different vegetable materials and natural fermentation methods influence the diverse physical, chemical, nutritional, and functional attributes of the products. All the traditionally fermented Thai vegetable samples collected showed physicochemical properties associated with the fermentation process, contributing to the nutritional and functional quality of the final products. Achieving consistent research results is challenging due to the intricate nature of food matrices and biochemical processes during fermentation. The roles of microorganisms, especially probiotics, are crucial in delivering health benefits through fermented foods. Traditionally fermented Thai vegetable foods contain high levels of total soluble solids, titratable acidity, and salinity in pickled shallot and ginger as a result of the natural fermentation process and the ingredients used. The research findings were confirmed using a hierarchical cluster analysis (HCA)-derived dendrogram pattern. The nutritional compositions, total phenolic contents, and antioxidant activities varied among the different types of vegetables. The correlations among lipid, protein, fiber, total soluble solid (TSSs), total titratable acidity (TTA), and salinity as potential biomarkers in fermented vegetable products were examined. The results suggest that traditionally fermented Thai vegetable products significantly impacted food research by enhancing the quality and preserving the authenticity of traditionally fermented Thai vegetables.

Astaxanthin is a red pigment that is naturally produced by the freshwater microalga Haematococcus pluvialis. Cell wall thickness of red cyst aplanospores is a barrier for extraction. Astaxanthin extraction from H. pluvialis UTEX 2505 was improved by ultrasonic assistance techniques with various conditions of solvent, extraction time and cycles of extraction to generate maximum recovery. Highest astaxanthin content and extraction yield were obtained using ethyl acetate under ultrasonic treatment for 25 min with five repeated extraction cycles. Encapsulation of astaxanthin was studied at different conditions. Ratio of maltodextrin and gum Arabic at 7.5:2.5 was most suitable for astaxanthin microencapsulation. Increased encapsulation efficiency of astaxanthin powder is suitable for food, pharmaceutical and other natural astaxanthin applications from H. pluvialis.

Irrational and injudicious use of antibiotics, easy availability of them as over-the-counter drugs in economically developing countries, and unavailability of regulatory policies governing antimicrobial use in agriculture, animals, and humans, has led to the development of multi-drug resistance (MDR) bacteria. The use of medicinal plants can be considered as an alternative, with a consequent impact on microbial resistance. We tested extracts of Piper longum fruits as new natural products as agents for reversing the resistance to antibiotics. Six crude extracts of P. longum fruits were utilized against a clinical isolate of multidrug-resistant Staphylococcus aureus. The antibiotic susceptibility testing disc method was used in the antibiotic resistance reversal analysis. Apart from cefoxitin and erythromycin, all other antibiotics used (lincosamides [clindamycin], quinolones [levofloxacin and ciprofloxacin], and aminoglycosides [amikacin and gentamicin]) were enhanced by P. longum extracts. The extracts that showed the greatest synergy with the antibiotics were EAPL (ethyl acetate [extract of] P. longum), n-BPL (n-butanol [extract of] P. longum), and MPL (methanolic [extract of] P. longum The results of this study suggest that P. longum extracts have the ability to increase the effectiveness of different classes of antibiotics and reverse their resistance. However, future studies are needed to elucidate the molecular mechanisms behind the synergy between antibiotic and phytocompound(s) and identify the active biomolecules of P. longum responsible for the synergy in S. aureus.

The β-carotene-rich yeast Rhodotorula paludigena CM33 was grown in a 22-L bioreactor using sugarcane molasses in combination with several distinct fed-batch strategies: DO (dissolved oxygen)-stat; stepwise feeding; and hybrid feeding (an initial exponential feeding phase followed by a DO-stat operation). The impacts of feeding strategies on biomass production, carotenoid production and nutrient conversion efficiencies were evaluated. The stepwise feeding proved most effective, achieving: a final dry biomass concentration of 73.1 ± 2.7 g L−1; a β-carotene titer of 181.9 ± 1.0 mg L−1; a biomass volumetric productivity of 1.33 ± 0.05 g L−1 h−1; and β-carotene productivity of 3.31 ± 0.02 mg L−1 h−1. However, with this feeding strategy the sugar and nitrogen conversion efficiencies were the lowest: the β-carotene yield on total sugar was 0.92 ± 0.00 mg g−1 and its yield on nitrogen was 5.47 ± 0.06 mg g−1. In contrast, the DO-stat operation was the most resource efficient, providing a biomass yield on sugar of 0.47 ± 0.02 g g−1, a biomass yield on nitrogen of 2.64 ± 0.05 g g−1, a β-carotene yield on sugar of 1.08 ± 0.04 mg g−1, and β-carotene yield on nitrogen of 6.08 ± 0.08 mg g−1. The hybrid strategy resulted in intermediate values of the various performance parameters. Raw sugarcane molasses proved to be excellent for high-cell-density culture of R. paludigena. A compromise between productivity and resource use efficiency may be required for commercial production of β-carotene especially if the substrate is expensive compared to the product.

In an effort to use baby corn husk (BCH) as a sustainable feedstock for cellulase and xylanase production by the Lentinus squarrosulus Mont. isolate LS-YA (LSM-LS-YA), a suitable pretreatment method and fermentation strategies were developed. BCH pretreated with 1 M sodium hydroxide for 90 min, an alkaline pretreatment, exemplified an appropriate pretreatment method. In a 10-L external Venturi injector bioreactor, the highest cellulase and xylanase production was 4.12 ± 0.36 unit/mL and 6.15 ± 0.36 unit/mL, respectively, when 1 g/L diammonium hydrogen phosphate was used as the nitrogen source and the aeration rate was controlled at 0.2 vvm. This study provides an informative perspective on the production of cellulase and xylanase from agricultural lignocellulosic materials, which could reduce agricultural waste while supporting a zero-waste circular economy, and this fermentation process would be applicable to larger-scale production.
