Item type:Publication, Hidden diversity of Pestalotiopsis and Neopestalotiopsis (Amphisphaeriales, Sporocadaceae) species allied with the stromata of entomopathogenic fungi in Taiwan(2024-01-01) ;Hsu, Sheng Yu ;Xu, Yuan Cheng ;Lin, Yu Chen ;Chuang, Wei YuLin, Shiou RueiPestalotiopsis sensu lato, commonly referred to as pestalotiopsis-like fungi, exhibit a broad distribution and are frequently found as endophytes, saprobes and pathogens across various plant hosts. The taxa within pestalotiopsis-like fungi are classified into three genera viz. Pestalotiopsis, Pseudopestalotiopsis and Neopestalotiopsis, based on the conidial colour of their median cells and multi-locus molecular phylogenies. In the course of a biodiversity investigation focusing on pestalotiopsis-like fungi, a total of 12 fungal strains were identified. These strains were found to be associated with stromata of Beauveria, Ophiocordyceps and Tolypocladium in various regions of Taiwan from 2018 to 2021. These strains were evaluated morphologically and multi-locus phylogenetic analyses of the ITS (internal transcribed spacer), tef1-α (translation elongation factor 1-α) and tub2 (beta-tubulin) gene regions were conducted for genotyping. The results revealed seven well-classified taxa and one tentative clade in Pestalotiopsis and Neopestalotiopsis. One novel species, Pestalotiopsis manyueyuanani and four new records, N. camelliae-oleiferae, N. haikouensis, P. chamaeropis and P. hispanica, were reported for the first time in Taiwan. In addition, P. formosana and an unclassified strain of Neopestalotiopsis were identified, based on similarities of phylogeny and morphology. However, the data obtained in the present study suggest that the currently recommended loci for species delimitation of pestalotiopsis-like fungi do not deliver reliable or adequate resolution of tree topologies. The in-vitro mycelial growth rates of selected strains from these taxa had an optimum temperature of 25 °C, but growth ceased at 5 °C and 35 °C, while all the strains grew faster under alkaline than acidic or neutral pH conditions. Item type:Publication, The family 36 carbohydrate-binding module of Paenibacillus xylaniclasticus TW1 xylanase: Characterization and recognition in epidermal tissue of sweet potato roots(2025-07-01); ; Paenibacillus xylaniclasticus TW1 was able to aerobically produce a multienzyme complex. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) of the crude enzyme preparation revealed at least 12 proteins that were bound to insoluble cellulose. Only the band representing cellulose-bound protein 12 (CBP12) from SDS-PAGE was identified as xylanase family 11 with a carbohydrate-binding module family 36 (CBM36) using matrix-assisted laser desorption/ionization time-of-flight/time-of-flight mass spectrometry (MALDI-TOF/TOF MS). In this report, CBM36 of P. xylaniclasticus TW1 (PxTW1CBM36) was cloned, expressed, purified and studied for binding characteristic. The results found that PxTW1CBM36 displayed broad binding ability to polysaccharides which high affinity for xylan and insoluble cellulose. Interestingly, this is the first report indicated that CBM36 had an affinity for insoluble cellulose. Although the amino acid residues involved in PxTW1CBM36 binding were conserved, the binding capacity of PxTW1CBM36 do not perturb by the addition of ethylenediaminetetraacetic acid (EDTA). It is possible that PxTW1CBM36 had different binding mechanisms with other CBM36. In addition, the binding characteristic of CBM36 on polysaccharides embedded within plant cell walls was also elucidated. It displayed the strong recognition for ligands located in epidermal tissue of sweet potato roots. Item type:Publication, Comparative evaluation of leaf and seed methanolic extracts obtained from Sophora tomentosa Linn. for phytochemical, phenolic content, antioxidant and antibacterial activities(2023-01-01) ;Laowklang. Poeaim, P. S.Sophora tomentosa Linn. (Fabaceae), also known as a yellow necklace pod, is a traditional medicinal plant widely used to treat dysentery and diarrhea. This study evaluated the biological activities of methanolic leaf and seed extracts of S. tomentosa. The total phenolic content of methanolic leave (STL) seed (STS) and dechlorophyll of leave extract (DSTL) showed 92.02 ±1.78, 38.48 ± 0.61, and 35.35 ± 2.45 mg GAE/g extract), respectively. For antioxidant activity, DPPH, ABTS and FRAP assay was found in leaves (STL) more than in dechlorophyll of leave extract (DSTL) and seed (STS). The methanolic extracts were assessed for antibacterial activity against eight pathogenic bacterial species. The seed extract showed the most potent antibacterial activity against Kocuria rhizophila with 12.85±3.85 mm inhibition zones. For phytochemical evaluation, both leaf and seed extracts exhibited alkaloids and coumarin. The results showed that the leaf extract (STL) had higher antioxidant activity and total phenolic content than seeds and dechlorophyll of leave extract (DSTL). On the other hand, the seed extract presented the most potent antibacterial activity. The current study suggested that both leave and seed extracts of S. tomentosa gave the potential source of natural bioactive compounds, which will be further evaluated in other biological activities. - Some of the metrics are blocked by yourconsent settings
Item type:Publication, Application of organic soybean tempeh in purple sweet potato raisin cookie(2022-05-01); Organic soybean tempeh or tempeh kedelai is made by fermentation with Rhizopus oligosporus was used to produce cookies, as a plant-based protein snack. Four samples with various recipes - purple sweet potato cookie, purple sweet potato raisin cookie, tempeh purple sweet potato cookie and tempeh purple sweet potato raisin cookie – were prepared to determine the physicochemical and sensory features. Chemical analysis found that the tempeh purple sweet potato raisin cookie has the highest moisture (18%) and total proteins (29%). However, the purple sweet potato cookies showed the highest ashes content (2%) and tempeh purple sweet potato cookie showed the highest total lipids (29%). The tempeh purple sweet potato raisin cookie was hardest (61 N). The tempeh purple sweet potato raisin cookie had the highest scores for food acceptability of all qualities in appearance, texture, aroma, colour, taste and general acceptability using a 9-point hedonic scale ranging from slightly liked to very much liked. Therefore, the tempeh purple sweet potato raisin cookie can be accepted as a new snack, that includes a good level of protein and thus a healthy food alternative. Furthermore, if all steps of the production are carefully controlled by USDA or other certifying bodies, thus this product may be labelled as organic product. - Some of the metrics are blocked by yourconsent settings

For morphology, both Cookeina sp. are white mycelium and the growth rate on potato dextrose agar (PDA) result present C. sulcipes is faster than C. tricholoma. the molecular characterization from a rapid and simple DNA extraction method that is modified based on thermolysis method, the DNA extraction is finish in thirty minutes and efficiency to continuous with polymerase chain reaction (PCR) amplification to fungi species level identification. the DNA sequence from internal transcribed spacer (its) gene regions by universal primer pairs its5/its4 is effective to confirm Cookeina species level that C. sulcipes has 616 bp and C. tricholoma has 570 bp. including, DNA sequence of large subunit (lsU) gene regions by universal primer pairs lROR/lR5 is generate that C. sulcipes has 912 bp and C. tricholoma has 906 bp. the cultures are screened for antibacterial activity by agar plug diffusion method and found that both isolates have been no activity against test strains (Bacillus subtilis, Escherichia coli, Kocuria rhizophila (Micrococcus luteus), Pseudomonas aeruginosa, Staphylococcus aureus and S. epidermidis). in a preliminary screening test of enzymes involved in plant biomass breakdown by agar plate method, both Cookeina sp. show cellulolytic and hemicellulolytic enzymatic activity, and manganese peroxidase (MnP) productivity. in contrast, only C. sulcipes had additional laccase activity. Item type:Publication, Diversity of Sordariales Fungi: Identification of Seven New Species of Naviculisporaceae Through Morphological Analyses and Genome Sequencing(2025-12-01); ;Gautier, Valérie ;Lalanne, Christophe ;Bonometti, LucasCros-Arteil, SandrineThanks to next-generation sequencing (NGS) technologies, the diversity of fungi can now be investigated through the analysis of their genome sequences. Naviculisporaceae is a family within the Sordariales, whose diversity is not well-known, with only one genome sequence published for this family. Here, we report on the isolation and cultivation of 20 new strains of Naviculisporaceae. Their genome sequences, as well as those of the five commercially available strains, were determined, thus providing complete genome sequences for 25 new Naviculisporaceae strains. Species delimitation was conducted using a combination of (1) ITS + LSU phylogenetic analysis of the new isolates along with other known species of the family, (2) comparisons between DNA barcode sequences of the new strains with those of the known species, and (3) average genome-wide nucleotide identity calculation. We built a phylogenomic tree and studied the organization of the mating-type locus. In vitro fruiting was obtained for 16 strains, enabling the definition of seven new species, namely Pseudorhypophila gallica, Pseudorhypophila guyanensis Rhypophila alpibus, Rhypophila brasiliensis, Rhypophila camarguensis, Rhypophila reunionensis and Rhypophila thailandica, as well as two new combinations, namely Pseudorhypophila latipes and Pseudorhypophila oryzae. Eight strains for which in vitro fruiting was not obtained may belong to additional new species. Niessl Species Complex in Metropolitan and Overseas France with Description of a New Species, Podospora reunionensis Silar, sp. nov.(2025-12-16) ;Silar, Philippe ;Gautier, Valérie ;Lalanne, Christophe; Arthur, MarvynPodospora anserina (Rabenh.) Niessl is a model fungus that was shown to belong to a complex of seven species with morphologically indistinguishable sexual fruiting bodies. Through the morphological analyses and ITS (Intergenic Transcribed Spacer) sequences of 86 newly-isolated strains, as well as the genome sequences of five strains, we show that members of the P. anserina species complex present different distribution ranges in metropolitan and overseas France. Podospora anserina is present all-over metropolitan France, including Corsica, while P. comata Milovtz. is restricted to the Northern part and P. pauciseta (Ces.) Traverso to the Southern part of continental France. The Guadeloupe hosts P. pseudoanserina C.Boucher, T.S.Nguyen & P.Silar and La Réunion island a species new to science, described here as Podospora reunionensis Silar, sp. nov. Item type:Publication, Evaluation of genetic diversity by molecular markers in Indian gooseberry (Phyllanthus emblica)(2025-01-01); ;Sangsoy, V.The genetic diversity and relationships among 16 cultivars of Indian gooseberry (Phyllanthus emblica) using Sequence-Related Amplified Polymorphism (SRAP) and Random Amplified Polymorphic DNA (RAPD) markers were investigated. The cultivars were collected from the Phrae Horticultural Research Center, Thailand. For SRAP analysis, 30 primer combinations were initially tested, comprising five forward and six reverse primers. Seven primer combinations consistently generated well-defined bands and were selected for genetic profiling. These SRAP primers amplified 172 fragments, with 145 (84.30%) polymorphic. For RAPD analysis, 50 primers were evaluated, and eight primers consistently produced clear bands of genetic profiling. These RAPD primers amplified 215 fragments, with 188 (87.44%) polymorphic. The genetic relationships among the cultivars were assessed using a dendrogram constructed with NTSYS-pc (version 2.1X) based on the Unweighted Pair Group Method with Arithmetic Mean (UPGMA). The genetic similarity coefficients ranged from 0.66 to 0.90 for SRAP and from 0.58 to 0.89 for RAPD markers, indicating a high level of genetic diversity among the cultivars. - Some of the metrics are blocked by yourconsent settings

P.The genetic diversity of 22 cassava varieties using the inter-simple sequence repeat (ISSR) molecular marker was investigated. Genetic similarity was derived from the matrix of simple similarity coefficients using the unweighted pair group method with arithmetic average (UPGMA). The genetic similarity coefficient ranged from 0.73 to 0.94. The similarity coefficient at 0.73, cluster analysis was clustered to be 22 cassava varieties into two significant groups. The first group was the sweet variety of cassava used for consumption, consisting of Rayong2, Hana-tee and variegated leaves used for decoration. Another group, the bitter variety of cassava used for industry, consisted of Rayong1, Rayong3, Rayong5, Rayong7, Rayong9, Rayong11, Rayong15, Rayong60, Rayong72, Rayong86-13, Rayong90, Kasetsart50, Kasetsart72, Huay Bong60, Huay Bong80, Huay Bong90, Pirun1, Pirun2 and unknown. The sweet and bitter variety contained different levels of cyanide content. JayaramaDai, Dong QinCoelomycete is a general term used for asexual fungi which produce conidia in fruiting bodies: pycnidial, acervular, cupulate, pycnothyria or stromatic conidiomata. The group contains numerous plant pathogenic, saprobic and endophytic species associated with a wide range of hosts. Traditionally, morphological characters and host associations have been used as criteria to identify and classify coelomycetes, and this has resulted in a poor understanding of their generic and species boundaries. DNA based taxonomic studies have provided a better outlook of the phylogenetic and evolutionary trends in coelomycetes. However, the present outcomes represent only a preliminary step towards the understanding of coelomycetes. Many genera have not been revisited since they were first described. The present study revises the classification of the hyaline-spored coelomycetes and provides a modern taxonomic framework based on both morphology and phylogeny. In total, 248 genera were investigated, of which less than 100 are known to have sequence data. Multi-locus sequence data analyses of 28S nrDNA, 18S nrDNA, ITS, RNA polymerase II second largest subunit (rpb2), and part of the translation elongation factor 1-alpha gene (tef1) and β-tubulin (tub2) gene regions were analysed. As a result, three new genera and 23 new species are introduced. In addition, three new links between sexual and asexual genera are provided. There are 138 genera that lack sequence data, and these are treated as Ascomycota, genera incertae sedis. Line drawings and descriptions are provided based on the examination of types and fresh collections and on the literature.
