Abstract
A successful protocol was established for callus induction and regeneration of Stemona curtisii Hook. f. The sterilized seeds were cultured on Murashige and Skoog (MS, 1962) supplemented with 0-0.5 mg L-1 2,4-dichlorophenoxyacetic acid (2,4-D) in dark and light conditions. The combination of 0-2 mg L-1 of 2,4-D and benzyladenine (BA) or thidiazuron (TDZ) was also examined in seed and shoot culture. The results show that callus induction was highest from seed. The 0.4 mg L-1 2,4-D in the dark condition treatment was suitable to promoted callus formation at 62.50%. The combination of 2,4-D with BA or TDZ strongly induced callus formation (2 mg L-1 BA with 2 mg L-1 2,4-D). Plant regeneration from callus was successfully induced via both organogenesis and embryogenesis.
