    Study on the regeneration of sausage tree (Kigelia africana) by tissue culture

    This study investigated the regeneration of sausage tree (Kigelia africana) using tissue culture techniques, focusing on callus induction, shoot regeneration from seeds, and root induction from shoot explants. Seeds were cultured on solid MS medium supplemented with various concentrations of BA (0.5, 1, 2, 3, and 5 mg L⁻¹) for 10 weeks. Callus formation was observed at all BA concentrations, with the largest mean callus volume (5214.17 mm³) and highest induction rate (80%) achieved at 5 mg L⁻¹. Shoot regeneration occurred across all BA treatments, with the highest number of shoots (45 shoots; 86.54%) obtained on medium containing 1 mg L⁻¹ BA. Root induction was studied using shoot segments derived from seed-induced shoots cultured on MS medium supplemented with IAA, NAA, or IBA for 4 weeks. IAA at 0.5 mg L⁻¹ induced roots in 33.33% of explants with an average root length of 6.68 mm. NAA at 2 mg L⁻¹ promoted the highest root induction (70.83%), though roots were shorter, while 1 mg L⁻¹ NAA produced fewer roots but with greater length (7.00 mm). IBA at 3 mg L⁻¹ induced rooting in 33.33% of explants with an average root length of 3.78 mm and also stimulated callus formation, with the largest callus volume observed at 2 mg L⁻¹. These results demonstrate that BA is effective for callus and shoot induction, whereas IAA, NAA, and IBA promote root formation, with distinct responses at different concentrations. The optimized protocols provide a foundation for in vitro propagation, mass multiplication, and conservation of K. africana, with potential applications in biotechnology and secondary metabolite production.

    The effect of plant growth regulator and in vitro conservation of teak (Tectona grandis L.) by tissue culture

    The plant regeneration of teak (Tectona grandis L.) was reported. Result showed that the highest number of shoot induction was 55.49 cm., in length of shoots which tested at 0.25 mg·L <sup>-1</sup> BAP, and averaged 40.29 cm which tested at 0.25 mg·L <sup>-1</sup> mT after 4 weeks. The shoots were elongated using shoot tip explants of cryopreservation on teak by encapsulation-dehydration (EnDe), encapsulation-vitrification (EnVi) and vitrification (Vi) techniques. The shoot tips were used as explants, encapsulated apices were precultured on hormone-free MS medium containing 0.3 M sucrose for 2 days, dehydrated, and immersed in liquid nitrogen for 24 h after that thawed in thermal bath at 40 ºC and washed in unloading solution. Vi and EnVi protocols were used to treat with a loading solution, dehydrated by plant vitrification solution 2 (PVS2). EnDe protocol were encapsulated by desiccated air-dry in laminar air flow and dehydrated with silica gel. Results showed that Vi gave an excellent shoot tip explants for survival and regrowth. Vi were unable to survival. This experiment was preliminary searched to improve and develop the teak for cryopreservation.

    Induction of shoot and root from nodes of Kadsura heteroclita

    Results described on shoot and root induction from node of medicinal plant Kadsura heteroclita. The sterilized nodes were used as explants and cultured on solid synthetic medium, Murashige and Skoog (MS) medium supplemented with single or combined Plant Growth Regulators (PGRs) of 6-benzylaminopurine (BAP), meta-Topolin (mT), Indole-3-butyric acid (IBA),1-Naphthaleneacetic acid (NAA) and Gibberellin (GA <inf>3</inf> ) were applied. The best result for shoot induction was shown on the medium supplemented with 0.5 mg/L of mT after 8 weeks. Transferred shoot to MS medium supplemented with combination PGRs of 0.5 mg/L of mT and 0.5, 1.0, 1.5 and 2.0 mg/L of IBA NAA or GA <inf>3</inf> after 16 weeks revealed that the explant were developed to multiple shoots which averaged number of shoot were 3 shoots/explant. The roots were not perfectly exhibited like sore on medium supplemented with 0.5 mg/L of mT and 1.5 mg/L of GA <inf>3</inf> . The high concentration of Auxin inhibited growth roots. The root induction was done by cutting the shoot and cultured on MS medium supplemented with combination PGRs of 0.5 mg/L of mT and 0.01, 0.02, 0.03, 0.04 and 0.05 mg/L of IBA after 4 weeks found that root length was approximately 2 mm on 0.5 mg/L of mT and 0.05 mg/L of IBA.

    Efficiency of antioxidant and absorbent on browning and the optimal factors of plant regeneration from young seed of gluta usitata (217 Mae Ka) by Tissue Culture

    Gluta usitata (Anacardiaceae) is a lacquer tree and create lacquer varnish can be utilized in many aspects especially art and culture. Recently, its harvest faced troublesome and yields of varnish tree are decreased. Consequently, the conservation of plant has reduced, and this plant has become extinct. The experiment was chosen to use plant tissue culture technique to preserve and increase this plant. The experiment was compared the effect of Woody Plant Medium (WPM) combined with different concentration of antioxidants were 2, 4, 6, 8 g/L Poly Vinyl Pyrrolidone (PVP), 20, 30, 40, 60 mg/L ascorbic acid or citric acid and absorbents of 1, 2, 3 g/L activated charcoal to reduce browning of phenolic compound in the explants. The results showed that 2 g/L activated charcoal gave the lowest of browning on seed and medium (about 0.9±0.2 points). The addition of 40 mg/L ascorbic acid was generated the highest of shoots (approximately 1.88±0.4 shoots/seed) after culture for 4 weeks. in vitro propagation of seed germination on WPM supplemented with 0.5, 1, 1.5, 2 and 3 mg/L 6-Benzyl amino purine (BAP), Thidiazuron (TDZ) and meta-Topolin (mT) 3% sucrose and 0.2% phytagel was efficiently resulted. Other result revealed that 1.5 mg/L BAP, 2 mg/L TDZ and 2 mg/L mT gave the highest seed germination at 1.6±0.2, 1.8±0.2 and 1.6±0.2 shoots/seed, respectively. The best of plant growth regulators for seed germination was 2 mg/L of TDZ after 6 weeks.

    Titanium dioxide (TiO2) nanoparticles induced callus induction and plant regeneration of indica rice cultivars (suphanburi1 and suphanburi90)

    Efficient plant regeneration systems are useful for indica rice using micropropagation and/or genetic transformation. Nanomaterials provide applications and opportunities in various fields of life science and agriculture technology. In this research, indica rice cultivars ‘Suphanburi1’ and ‘Suphanburi90’ (Oryza sativa L. ssp. indica) were used to investigate. Seeds were examined to evaluate callus induction and plant regeneration frequency. Culture of sterilized-seeds on callus induction medium supplemented with 50 mg L<sup>-1</sup> of TiO<inf>2</inf> nanoparticles induced a high frequency of callus induction and content of biomass accumulation. Embryogenic calli were transferred to plant regeneration medium supplemented with 10-50 mg L<sup>-1</sup> TiO<inf>2</inf> nanoparticles. The highest frequency of plant regeneration and the ratio of the number of seedlings to the number of regenerated calli were produced on plant regeneration medium supplemented with 40 mg L<sup>-1</sup> of TiO<inf>2</inf> nanoparticles. The regenerated shoots induced roots on NB medium without plant growth regulators and the plantlets were established in soil. The results may be used for nanomaterial applications of rice and/or other plants for its further improvement.

    Application of activated charcoal and nanocarbon to callus induction and plant regeneration in aromatic rice (Oryza sativa L.)

    The investigations of nanotechnology with the application on agricultural products also have been few reported, especially the plant regeneration. The effects of activated charcoal and nanocarbon on the callus induction and plant regeneration of aromatic rice were studied. Activated charcoal was added into the callus induction and regeneration medium. The presence of activated charcoal in the callus induction medium (100-500 mg L<sup>−1</sup>), activated charcoal significantly reduced the percentage of the callus induction and biomass accumulation (fresh weight, dry weight and size). Whereas, the regeneration medium supplemented with 100 mg L<sup>−1</sup> of activated charcoal showed the highest percentage of plant regeneration (61.90%) and the ratio of the number of seedlings to the number of regenerated calli (RSR; 3.06) that derived from the callus induction medium (without activated charcoal). Moreover, the induced calli derived from the callus induction medium supplemented with nanocarbon at 5 mg L<sup>−1</sup> showed the highest percentage of callus induction (94.70%), the percentage of green spots (95.83%), the percentage of plant regeneration (60.42%) and the RSR (3.12) when transferred the calli into the regeneration medium (without nanocarbon). After that, nanocarbon was also added into the regeneration medium. The percentage of green spots (96.08%), the percentage of plant regeneration (62.75%) and the RSR (3.16) obtained from the regeneration medium supplemented with 20 mg L<sup>−1</sup> of nanocarbon showed the highest values. This experiment showed that the optimum concentration of activated charcoal and nanocarbon had potential to enhance the callus induction and plant regeneration frequencies in tissue culture medium of aromatic rice.

    Application of nanomaterials in plant regeneration of rice (Oryza sativa L.)

    In recent years, the application of nanomaterials has been investigated by various studies, especially in their biological and agricultural features. This research aimed to evaluate the influence of zinc oxide (ZnO) and titanium dioxide (TiO<inf>2</inf>) nanoparticles on the quality of the plant regeneration in rice (Oryza sativa L. cv. RD49). Mature seed explants were used for callus induction and plant regeneration cultured on the medium supplemented with different concentrations of ZnO or TiO<inf>2</inf> nanoparticles. The high percentage of the callus induction was achieved on the medium supplemented with 5 mg.L<sup>-1</sup> ZnO nanoparticles or 20 mg.L<sup>-1</sup> TiO<inf>2</inf> nanoparticles when compared with control treatment (without nanoparticles). Various concentrations of TiO<inf>2</inf> nanoparticles were also found to increase the percentage of the plant regeneration more than ZnO nanoparticles. The highest percentage of plant regeneration was observed on the medium containing the concentration of 20 mg.L<sup>-1</sup> TiO<inf>2</inf> nanoparticles. The in vitro rooting of regenerated shoots was hardened and successfully transplanted to the field after 30 days of transplantation. The results suggested that ZnO and TiO<inf>2</inf> nanomaterials may have potential applications in the in vitro plant regeneration.

    A proper concentration of carbon black nanoparticles enhances growth of the regenerated vetiver grass

    Carbon black (CB) nanoparticles were used in enhancing efficiency of the plant regeneration process under tissue culture condition. The calli of vetiver grass (Vetiveria zizanioides L. Nash) were induced by using axillary bud as the explant. These calli were subjected onto plant regeneration medium, Murashige & Skoog (MS) medium supplemented with 1 mgL<sup>-1</sup> α-naphthalene acetic acid (NAA), 2 mgL<sup>-1</sup> 6-benzylaminopurine (BAP) and 2 mgL<sup>-1</sup> kinetin, adding 0-70 mgL<sup>-1</sup> concentrations of carbon black (CB) nanoparticles. After six week incubation, vetiver calli developed on the medium supplemented with 40 mgL<sup>-1</sup> CB nanoparticles showed the rather high frequencies of plant regeneration (93.75%), nevertheless no statistically different with other treatment did. Media containing CB nanoparticles in the range 40-60 mgL<sup>-1</sup> significantly affected on enhancing the average of shoot number per callus more than other CB concentration did. Especially, the average shoot lengths of plantlets derived from 40 and 50 mgL<sup>-1</sup> CB media (1.73 and 2.19 cm) are longer than those from non CB or other CB concentration media (0.88<sup>-1</sup>.05 cm) at the significant level. Adding CB nanoparticles in regeneration medium effects on enhancing electrical conductivity and decreasing the pH value. The concentration of CB nanoparticle effected the ratio of colloidal sizes which was found in the medium. The proper concentration of CB generates the optimum property of the regeneration medium which can promote the average of shoot number per callus, and enhanced shoot elongation of vetiver grass in the regeneration process.

    Establishment of condition and nanoparticle factors influencing plant regeneration from aromatic rice (Oryza sativa)

    Improvements on the callus induction and regeneration stages as well as the applications of nanoparticles using aromatic rice cv. KDML105 were investigated in this study. An efficient induction of plant regeneration from mature seeds was found in calli cultured under dark condition and was not transferred to the fresh medium (non-subculture) in each culture stage. Although, the size and biomass factors showed the better performance in calli cultured under light condition than dark but the regeneration frequency rate was lower than dark condition. Between the types and concentrations of nanoparticles applied for the regeneration enhancement. ZnO nanoparticles were found to be toxic, while TiO2 nanoparticles improved the plant regeneration. Optimum concentration (25 mg L<sup>-1</sup>) of TiO<inf>2</inf> nanoparticles showed the high rate of green spots, plant regeneration and the ratio of the number of seedlings to the number of regenerated calli. Nonetheless, these factors may be valuable to establish an improvement in plant tissue culture system for propagation and/or genetic transformation in aromatic rice.

    Plant regeneration from immature inflorescence of zoysiagrass (Zoysia spp.)

    Zoysiagrass displays good adaptability to its environment, and is utilized for golf courses and athletic fields. Immature inflorescences were obtained from 5 varieties of zoysiagrass, 'Miyako', 'Misato', 'Meyer', 'Yamato' and 'B14'. However, regenerated plants were only obtained in 'Miyako'. A relationship between the size of the inoculated inflorescences and the callus induction frequency was observed. A high frequency of callus formation was shown in earlier developmental stages shorter than 20 mm in the length of inflorescences in 'Miyako'. Plant regeneration was observed efficiently when the compact calli, which were formed in the callus induction medium (Linsmaier and Skoog 4 mg l<sup>-1</sup> thiamine-HCl, 100 mg l<sup>-1</sup> α-ketoglutaric acid, 2 mg l<sup>-1</sup> 2,4-D, 0.1 mg l<sup>-1</sup> BA, 3% sucrose, 0.2% gelrite), were transferred to the regeneration medium (Linsmaier and Skoog 1 mg l <sup>-1</sup> BA, 3% maltose, 0.5% gelrite) in 'Miyako'.
