    MYCO-MEDIATED SYNTHESIS AND α-GLUCOSIDASE INHIBITORY ACTIVITY OF SILVER NANOPARTICLES PRODUCED BY XYLARIACEOUS FUNGI

    Metal nanomaterials could be applied in various fields and could be synthesized via living organisms such as plants, bacteria and fungi. Here, the ability of Xylaria sp.5 to produce silver nanoparticles (AgNPs) and α-glucosidase inhibitory activities by the AgNPs produced by Xylaria sp.5 were investigated. The culture broth of Xylaria sp.5 was used to synthesize AgNPs by using 0.1 M of silver nitrate (AgNO3) solutions. The mycogenic crystals were investigated for the morphological characteristics and chemical composition by scanning electron microscope (SEM) equipped with energy dispersive X-ray spectroscopy (EDS), Fourier-transform infrared spectroscopy (FTIR) and X-ray powder diffraction (XRD). The results showed that AgNPs were successfully produced. Moreover, AgNPs were tested for α-glucosidase inhibitory activities. The crude enzyme derived from rat intestine consisted of maltase and sucrase. The percentage of inhibition at 50 mg/ml of maltase and sucrase was 63.21 ± 0.67 and 54.42 ± 0.11, respectively. This study demonstrated that the supernatant culture broth of Xylaria sp.5 can be used to synthesize AgNPs which possess α-glucosidase inhibitory activities. Collectively, this method could be a promising alternative for low cost and non-polluting production of AgNPs which could be potentially utilized for the treatment of type 2 diabetes mellitus.

    Differential responses to nanoparticles induced by nanocarbon application in pigmented rice seedlings

    To investigate the effects of nanocarbon on physiological responses of plant growth, contents of photosynthetic pigments, and the activities of antioxidant enzymes in the seedlings of pigmented rice cv. Hom-nin that were cultured on either untreated-nanocarbon (control) or treated-nanocarbon (soil:nanocarbon, 8:1, 6:1, 2:1 w/w) were studied. The results showed that growth performances, photosynthetic pigment contents and antioxidant enzyme activities in seedlings 7-28 days after the treated-nanocarbon were significantly differenced when compared to the control treatment. Growth performances, photosynthetic pigment contents and antioxidant enzyme activities in pigmented rice seedlings were significantly enhanced by the low and moderate concentrations of nanocarbon treatments (soil:nanocarbon, 8:1, 6:1 w/w) for all sampling dates (7, 14, 21 and 28 days after the nanocarbon treatments). While at the high concentration of nanocarbon (soil:nanocarbon, 2:1 w/w) treatment, growth performances, photosynthetic pigment contents and antioxidant enzyme activities in pigmented rice seedlings were significantly decreased when compared to the other treatments. The results suggested that the inducible physiological responses in pigmented rice seedlings to nanocarbon depend on both the concentrations and the exposure duration. The appropriate ratio of 8:1 and 6:1 w/w of soil:nanocarbon seemed to induce the physiological performances in the seedlings, more than those at the high concentration and untreated of nanocarbon.

    Enhancements of growth and metabolites of indica rice callus (Oryza sativa l. cv. pathumthani1) using tio2 nanoparticles (nano-tio2)

    In this research, the effects of a various in concentration of TiO<inf>2</inf> nanoparticles (Nano-TiO<inf>2</inf>) were applied to study the cell growth, metabolites and antioxidant responses of callus in indica rice plants. Dose-dependent changes in growth performances, metabolite accumulations and antioxidant activities of callus were found. Mature seeds of indica rice cv. Pathumthani1 were cultured in callus induction medium with different concentrations of Nano-TiO<inf>2</inf> (0-600 mg L<sup>-1</sup>). After five weeks of cultivation, the callus formations were measured in fresh weights and dry weights, and the contents of phenolic compounds, flavonoids, and the antioxidant activities of callus extracts were assayed. The Nano-TiO<inf>2</inf> induced of fresh weights and dry weights in callus formations when compared with the control treatment (0 mg L<sup>-1</sup> Nano-TiO<inf>2</inf>) were also observed. The flavonoid and phenolic compound contents in rice callus exposed to 400 mg L<sup>-1</sup> Nano-TiO<inf>2</inf> were 118% and 139%, respectively, higher than rice callus unexposed to Nano-TiO<inf>2</inf>. Moreover, the antioxidant activities of the callus metabolites studied shows that the percentages of inhibition were up-regulated (84%) when exposed to 400 mg L<sup>-1</sup> Nano-TiO<inf>2</inf>, as a response to the high-level depletion of the free radicals in metabolites of rice callus. The results obtained from this research showed that the addition of Nano-TiO<inf>2</inf> had impacts on the callus growth and metabolite accumulations in rice callus. The results suggested that addition of Nano-TiO<inf>2</inf> at appropriate levels could role as an elicitor for biosynthesis of valuable metabolites and antioxidant properties for pharmaceutical applications in further research.

    Biodiversity and bioactivity of stromatic xylariales collected from Mu Ko Chang National Park, Thailand

    Ko Chang Island is a tropical rain forest island of Mu Ko Chang national park, Thailand, which has a wide variety of macro-fungi. The objective of this research was to study the biodiversity of Xylariales and to investigate the bioactive activity of ethyl acetate crude extracts. The fungi were collected from the forest area of the waterfalls all over the island. The crude extracts of selected fungi were tested for antibacterial activity by agar well diffusion method and the cancer cell viability was assessed using the MTT assay. Three genera of Xylariaceae and Hypoxylaceae were found including Xylaria, Daldinia and Hypoxylon. The crude extracts inhibited the growth of Gram positive bacteria more than that of Gram negative bacteria and showed broad spectrum inhibition to all cancer cell lines. The crude extract of Xylaria sp.3 showed the highest potential of antibacterial activity and selectively inhibited gastric carcinoma (Kato III) and T cell leukemia (Jurket) with the percentage survival of 26.12 ± 0.90% and 35.37 ± 0.40% respectively. The molecular identification of Xylaria sp.3 was identified as Xylaria sp.TP5BS101. These results show that these macromycetes are an excellent repertoire of antibacterial activity and exhibit anticancer activity against Kato III and Jurket by inducing DNA fragmentation.

    Titanium dioxide (TiO2) nanoparticles induced callus induction and plant regeneration of indica rice cultivars (suphanburi1 and suphanburi90)

    Efficient plant regeneration systems are useful for indica rice using micropropagation and/or genetic transformation. Nanomaterials provide applications and opportunities in various fields of life science and agriculture technology. In this research, indica rice cultivars ‘Suphanburi1’ and ‘Suphanburi90’ (Oryza sativa L. ssp. indica) were used to investigate. Seeds were examined to evaluate callus induction and plant regeneration frequency. Culture of sterilized-seeds on callus induction medium supplemented with 50 mg L<sup>-1</sup> of TiO<inf>2</inf> nanoparticles induced a high frequency of callus induction and content of biomass accumulation. Embryogenic calli were transferred to plant regeneration medium supplemented with 10-50 mg L<sup>-1</sup> TiO<inf>2</inf> nanoparticles. The highest frequency of plant regeneration and the ratio of the number of seedlings to the number of regenerated calli were produced on plant regeneration medium supplemented with 40 mg L<sup>-1</sup> of TiO<inf>2</inf> nanoparticles. The regenerated shoots induced roots on NB medium without plant growth regulators and the plantlets were established in soil. The results may be used for nanomaterial applications of rice and/or other plants for its further improvement.
