Item type:Publication, Effects of organic compound and plant growth regulators on seed germination and growth of Doritis pulcherrima Lindl. in vitro(2024-09-01) ;Supreeworakij, S.; ;Rungkitwattananukul, T. ;Phanchompoo, P.Doritis pulcherrima Lindl. is one of the wild orchids of which the distribution has decreased in nature and endangered. In vitro seed culture by using plant tissue culture is a proper method to help increasing the numbers. The purpose of this research was to study the effect of organic compound and plant growth regulators on seed germination and growth of D. pulcherrima in vitro. Seeds were cultured on Vacin and Went (VW) basal medium. Experiments were performed in a 5×7 factorial in completely randomized design (factorial in CRD) 2 factors; Factor 1 capsule of D. pulcherrima with 5 treatments such as Pi102/1, Pi102/2, Pi119/1, Pi119/2 and Pu112. Factor 2 organic compound with 7 treatments such as no organic compound (control), 50, 100 g L-1 raw namwa banana powder, 50, 100 g L-1 cavendish banana homogenate, 50, 100 g L-1 potato homogenate. After 13 weeks of culture, there were only 3 capsules growth seed germination (Pi119/2, Pi102/2 and Pu112/1) and the highest percentage of seed germination was Pi119/2 on VW medium supplemented with 50, 100 g L-1 banana powder and 50 g L-1 potato homogenate gave the highest germination percentage at level 4 (75-100%). From the results, organic compound is a suitable culture supplement for D. pulcherrima seed germination and development compared to no organic compound. In addition, the effect of plant growth regulators on growth and development of D. pulcherrima protocorm were cultured on VW medium supplemented with different concentrations of control, 1, 2 and 3 mg L-1 BA and 0.01, 0.02 and 0.03 mg L-1TDZ. The result showed the control gave the highest number of leaves at 6.50, the number of protocorm at 3.67 and fresh weight of protocorm at 0.42 g. The protocorm and plantlet were induced after 10 of culture. - Some of the metrics are blocked by yourconsent settings
Item type:Publication, Mulberroside A Accumulation in Mulberry Plantlets from In Vitro Culture and Hydroponic System(2023-12-29) ;Damrongmongcolkul, Natvara; Lohasupthawee, PanaIn this study, nodal explants cultured in MS medium supplemented with BAP 4mg/l and NAA 0.5 mg/l induced 3.33 shoots per explant within 4 weeks. The elongated multiple shoots were transferred to rooting media. The best root development was found on MS medium without PGRs within 10 days. The acclimatized plants were transferred to a deep-water hydroponic system with different concentrations of MS nutrient solution without sucrose addition. The results showed that mulberry plants cultivated in 1/8 MS nutrient solution gave the highest growth index. For mulberroside A determination, the results showed that mulberroside A accumulation in roots were higher than in shoots of mulberry plants both for in vitro culture and in the hydroponic system. In the case of in vitro culture, mulberroside A accumulation in roots was 20.61 mg/g DW, and in shoots was 11.98 mg/g DW. In hydroponic system, mulberroside A accumulation in roots was 17.58 mg/g DW, and in shoots was 2.34 mg/g DW. We also found that the higher concentration of MS nutrient solution, the more mulberroside A accumulation in roots of mulberry plants cultivated in the hydroponic system. - Some of the metrics are blocked by yourconsent settings
Item type:Publication, The effect of auxins on root production and stemona alkaloid accumulation in Stemona curtisii Hook.f. in vitro culture(2023-02-01); ;Saenpakdee, S.Deewatthanawong, R.The herb Stemona curtisii Hook.f. has insecticidal properties. Its roots contain a specific group of stemona alkaloids. This study evaluated the effect of auxins on root production and stemona alkaloid accumulation in roots of S. curtisii. In vitro shoots were cultured on MS medium (Murashige and Skoog, 1962) supplemented with various types of auxins as indole-3-acetic acid (IAA), indole-3-butyric acid (IBA) and α-naphthalene acetic acid (NAA) at concentrations of 0, 0.2, 0.4, 0.6, 0.8 and 1.0 mg L-1 for 2 months. Results showed that auxins enhanced both fresh and dry root yields and also improved alkaloid contents compared to non-treated roots. Adding 0.6 mg L-1 IBA achieved highest root yield at 19.50% and stemocurtisinol content at 23.831×10-6 mg g-1 DW, while the highest total alkaloids and stemocurtisine contents were found in 0.8 mg L-1 IBA treatment at 186.976×10-3 mg g-1 DW and 340.142×10-6 mg g-1 DW, respectively.4
