    Investigation of Biodegradation and Biocompatibility of Chitosan–Bacterial Cellulose Composite Scaffold for Bone Tissue Engineering Applications

    Developing scaffolds with a three-dimensional porous structure and adequate mechanical properties remains a key challenge in tissue engineering of bone. These scaffolds must be biocompatible and biodegradable to effectively support osteoblastic cell attachment, metabolic activity, and differentiation. This study successfully fabricated a chitosan–bacterial cellulose (CS–BC) composite scaffold using the solvent casting/particle leaching (SCPL) technique, with NaOH/urea solution and sodium chloride crystals as the porogen. The scaffold exhibited a well-distributed porous network with pore sizes ranging from 300 to 500 µm. Biodegradation tests in PBS containing lysozyme revealed a continuous degradation process, while in vitro studies with MC3T3-E1 cells (pre-osteoblastic mouse cell line) demonstrated excellent cell attachment, as observed through SEM imaging. The scaffold also promoted increased metabolic activity (OD values) in the MTT assay, and enhanced alkaline phosphatase (ALP) activity and upregulated expression of osteogenic-related genes. These findings suggest that the CS–BC composite scaffold, fabricated using the SCPL method, holds great potential as a candidate for bone tissue engineering applications.

    A new nanofibre derived from Trichoderma hamatum K01 to control durian rot caused by Phytophthora palmivora

    Phytophthora rot of durian var Monthong caused by Phytophthora palmivora has been proven to be a serious threat to durian plantations in Thailand. The research was targeted to isolate the causal pathogen and prove its pathogenicity by Koch’s postulate. Morphology and molecular phylogeny have confirmed the identification of pathogenic and antagonistic fungi. Evaluation of antagonistic fungus against plant pathogen in vitro and greenhouse conditions, morphology and molecular phylogenetic identification confirmed antagonistic species Trichoderma hamatum K01 and pathogenic isolate P. palmivora PYSC01. The crude metabolite of T. hamatum K01, namely TK01-MeOH gave the most substantial inhibitory effect to inhibit colony growth and sporangia formation at ED<inf>50</inf> (50% effective dose) values of 288 and 118 µg/mL, respectively. Moreover, nanofibre namely nano-TK01M exhibited the best antifungal activity in inhibiting colony growth and sporangia formation at ED<inf>50</inf> values of 11 and 3 µg/mL, respectively. Nano-TK01M treated on durian leaves induced the synthesis of scopoletin, which is known as a defense mechanism and marker of plant resistance or plant immunity. Moreover, the application of nano-TK01M significantly reduced disease incidence, the same as metalaxyl. Additionally, nano-TK01M treatment was the most effective in enhancing plant physiological parameters, including the synthesis of chlorophyll, carotenoid contents and promoted plant growth, compared to both metalaxyl and non-treated control. T. hamatum K01 produced antifungal metabolite pyrone 6-pentyl-2H-Pyran-2-one and sorbicillin. It is reported for the first time that pyrone and sorbicillin could be expressed as bioactive compounds in reduction of the disease incidence of durian rot caused by P. palmivora. The finding confirmed that nano-TK01M from T. hamatum K01 exhibited the most effective in controlling plant pathogen, which could be promoted as agricultural input for plant disease management, and it is also a nontoxic fungicide for living life and eco-friendly.

    Four new species of genus Acmella W.T. Blanford, 1869 (Gastropoda, Assimineidae) from Southern Thailand

    This study explores the diversity of microsnails inhabiting limestone caves in Southern Thailand. It describes four new species of the genus Acmella W.T. Blanford, 1869 (Gastropoda, Assimineidae): Acmella krueangensis sp. nov. from Ranong Province, A. thamsingensis sp. nov. and A. changphueakensis sp. nov. from Chumphon Province, and A. kanchanaditensis sp. nov. from Surat Thani Province. These species are distinguished by shell morphology, particularly the number of striae on the last whorl and the protoconch sculpture. Phylogenetic analyses based on mitochondrial cytochrome c oxidase subunit I (COI) gene sequences further support their distinctiveness and clarify their taxonomic placement. The results enhance the understanding of Acmella diversity in Thailand and shed light on biogeographical patterns found in this genus and the adaptations needed to survive under the conditions of cave systems.

    Plant regeneration of Bauhinia purpurea by tissue culture technique

    Results showed that the seed of Bauhinia purpurea germination was archived 100% from half-strength MS medium supplemented with 3.0 mg/L Gibberellic acids (GA3) and gave 71.137 mm for the highest seedling. The optimal condition of nodal explants sterilization was E5, which gave 92% of survival explants. For the shoot induction method, the results showed that 24.270 mm was the highest proliferation of shoots achieved from MS medium supplemented with 1.0 mg/L 6-benzylaminopurine (BAP). The highest number of roots was 2.867 roots, and the highest root length was 33.453 mm, obtained at half-strength MS medium complemented with 0.25 mg/L Indole-3-acetic acid (IAA). The 73.33% of plantlets were survived after being transferred to plastic pot.

    New Pyrrolobenzoxazine Sesquiterpenoid Derivatives from the Fungus Talaromyces trachyspermus

    Three new pyrrolobenzoxazine sesquiterpenoids, talatrachyoxazines A-C (1-3), together with fourteen known compounds (4-17), were isolated from the fungus Talaromyces trachyspermus EU23. Their structures were identified by spectroscopic evidence and mass spectrometry. The absolute configurations of 1-3 were determined by NOESY data and comparison of their calculated and experimental electronic circular dichroism (ECD) spectra. Compound 1 showed cytotoxic activity against HelaS3, KB, HT-29, MCF-7, and HepG2 cell lines with IC50 values of 7, 11, 10, 12, and 10 μM, respectively. Compounds 1 and 14 showed weak antibacterial activity against the gram-positive bacteria Bacillus cereus and Bacillus subtilis, while 1-3 and 14 showed weak antibacterial activity against the gram-negative bacterium Pseudomonas aeruginosa. In addition, compound 1 showed weak antibacterial activity against Escherichia coli.

    Chemosensory-Driven Foraging and Nocturnal Activity in the Freshwater Snail Rivomarginella morrisoni (Gastropoda, Marginellidae): A Laboratory-Based Study

    Rivomarginella morrisoni is a freshwater snail endemic to Thailand, yet its behavioral ecology remains poorly understood. This study described the feeding behavior of R. morrisoni, focusing on its foraging activity, behavioral patterns, and food detection mechanisms under laboratory conditions using specimens collected from four river basins in central Thailand. Daily monitoring revealed nocturnal emergence, peaking between 21:00 and 22:00 h, with stable rhythms established 72 h post-feeding. Feeding trials revealed a preference for aged shrimp over fresh or decayed ones. Behavioral observations confirmed that food localization in R. morrisoni was mediated by chemical cues. Light–dark tests indicated a slight tendency toward darkness, but no significant phototactic response was observed. These findings suggest that R. morrisoni is a generalist scavenger with chemosensory-driven foraging and nocturnal activity. Its apparent sensitivity to habitat disturbance underscores the relevance of behavioral studies for informing future conservation and captive breeding efforts.

    Identification of SRAP and AFLP molecular markers associated with fruit traits in santol (Sandoricum koetjape)

    The genetic diversity of santol (Sandoricum koetjape) using sequence-related amplified polymorphism (SRAP) and amplified fragment length polymorphism (AFLP) markers was investigated. Both molecular markers showed associations between fruit quality traits of economic importance, such as fruit weight and sweetness. The SRAP results revealed 128 amplified bands across nine primer pairs, indicating a polymorphism rate of 41.41%. Moreover, utilizing a genetic similarity coefficient of 0.85, santol cultivars were categorized into two major groups. Additionally, the AFLP analysis identified 171 amplified bands from seven primer pairs with a genetic similarity coefficient of 0.79. Consequently, santol cultivars were further classified into three distinct groups based on these findings. When combining the SRAP and AFLP results, a total of 299 amplified bands were analyzed, resulting in the separation of santol cultivars into four groups. These findings demonstrated that using SRAP and AFLP, as well as their combined results, could elucidate the genetic diversity within santol cultivars and may contribute to their classification.

    Influence of leaf type, genotype, and location on bioactivities and assessment of genetic diversity in teak (Tectona grandis L. f.)

    Marked variation in biological activities was observed among teak (Tectona grandis L. f.) leaf extracts as influenced by leaf type, tree genotype, and planting location. Fresh leaves generally exhibited stronger antibacterial activity than fallen leaves, with significant differences detected among plus trees originating from Chiang Mai, Phrae, Sukhothai, Lampang, and Khon Kaen and grown at the Thongphaphum and Phitsanulok Silviculture Research Stations. Among all samples, the Phrae plus tree cultivated at Thongphaphum showed the highest antibacterial inhibition against the tested microorganisms. Methanol extracts demonstrated antibacterial, anti-tyrosinase, and anti-inflammatory activities, while sequential extraction of Phrae plus tree leaves using hexane and dichloromethane yielded fractions with potent antibacterial effects, indicating promising potential for the development of new antibacterial agents. Genetic analysis based on sequence-related amplified polymorphism (SRAP) markers revealed similarity coefficients ranging from 0.67 to 1.00, reflecting low to moderate genetic diversity among the studied germplasm, with some samples exhibiting nearly identical genetic profiles. Overall, the observed variation in biological activities was closely associated with both genetic background and environmental conditions, providing valuable information for teak germplasm selection and the utilization of leaf-derived bioactive compounds in product development.

    Application of organic soybean tempeh in purple sweet potato raisin cookie

    Organic soybean tempeh or tempeh kedelai is made by fermentation with Rhizopus oligosporus was used to produce cookies, as a plant-based protein snack. Four samples with various recipes - purple sweet potato cookie, purple sweet potato raisin cookie, tempeh purple sweet potato cookie and tempeh purple sweet potato raisin cookie – were prepared to determine the physicochemical and sensory features. Chemical analysis found that the tempeh purple sweet potato raisin cookie has the highest moisture (18%) and total proteins (29%). However, the purple sweet potato cookies showed the highest ashes content (2%) and tempeh purple sweet potato cookie showed the highest total lipids (29%). The tempeh purple sweet potato raisin cookie was hardest (61 N). The tempeh purple sweet potato raisin cookie had the highest scores for food acceptability of all qualities in appearance, texture, aroma, colour, taste and general acceptability using a 9-point hedonic scale ranging from slightly liked to very much liked. Therefore, the tempeh purple sweet potato raisin cookie can be accepted as a new snack, that includes a good level of protein and thus a healthy food alternative. Furthermore, if all steps of the production are carefully controlled by USDA or other certifying bodies, thus this product may be labelled as organic product.

    Identification of phenolic compounds and evaluation of biological activities of methanolic extracts obtained from two varieties of longan (Dimocarpus longan) peels

    Dimocarpus longan belongs to the Sapindaceae family, which are revealed as cryptic species. In Thailand, Dimocarpus longan spp. longan var. longan such as Edor is the most cultivated. Another Dimocarpus longan spp. longan var. obtusus (Dimocarpus obtusus), called Lum-Yai Thao, is found in Eastern Thailand. However, there is still little information on the biological activity of Thao. Hence, this research was designed to identify phenolic compounds and evaluate the biological activities of methanolic extracts obtained from peels of two longan varieties: Edor VS Thao. For the total phenolic compound, no significant differences were observed between Edor and Thao. For antioxidant activity using DPPH, ABTS, and FRAP assays, the analogs showed Edor had more activity than Thao. The disc diffusion test at 5 mg/disc showed that Edor peels extract effectively inhibited the growth of bacteria, but not Thao extracts which were strong against Propionibacterium acnes. However, both of them also did not have activity against Escherichia coli and Staphylococcus epidermidis.For anti-tyrosinase activity, both variety extracts had less effective anti-tyrosinase activity. In addition, anti-inflammatory activity was evaluated by measuring nitric oxide (NO) generated from SNP and using LPSstimulated RAW 264.7 cells which showed excellent activity.In cytotoxicity using MTT assay, the results demonstrated that the methanolic extract of longan peel from Thao exhibited low cytotoxicity against both L929 and HaCaT. On the other hand, the extract from Edor exhibited cytotoxic activity. These results suggested that Thao peel extracts are different from Edor peel extract. Thao peel extracts showed excellent anti-inflammatory and little cytotoxic activity for L929 and HaCaT cell lines and antibacterial against P. acnes. Therefore, Thao peel extracts could be used in preparing products for antiinflammatory supplements and cosmetics.
