The concentration of 1.5 mg/L of mT showed the highest average length of shoots which was 9.82 mm, while the 0.5 mg/L mT revealed the most significant number of roots which were 0.9 roots per explant with the longest root of 3.75 mm. The liquid Gamborg's B5 medium showed the highest average number of shoots at 1.5 mg/L BAP, the same as at 1.5 mg/L mT. The medium without plant growth regulator and 1 mg/L mT was the highest average length of shoots of 13.98 mm. The highest number of roots per explant was 0.8, similar to 2 mg/L mT, but the different maximum length of roots of 3.04 mm. A hundred percent of protocorms induced shoots found in all media. The plantlets were survived after transplanting into pots and covered with plastic bags in four weeks. - Some of the metrics are blocked by yourconsent settings
Item type:Publication, The effects of abscisic acid and sorbitol on the slow growth in vitro preservation of Mahesak (Tectona grandis L.)(2020-09-01) ;Tongsad, P. ;Laipasu, P. ;Chareonsap, P. P.Poeaim, AnuragThe conservation of teak (Tectona grandis L.) by slow growth was proceeded out to determine appropriate methods in vitro preservation using osmotically active compounds and growth retardants. Calli and node explants were cultured on Murashige and Skoog (MS) and Woody Plant Medium (WPM) media supplemented with 1, 2 and 3 mg. L-1abscisic acid (ABA) containing 30 g.L-1sucrose and hormone-free MS and WPM media containing 0.1, 0.2 and 0.3 M sorbitol for reduced the growth rate. Result showed that preservation of calli on MS and WPM media supplemented with 0.2 and 0.3 M sorbitol and node explants were cultured on WPM medium supplemented with 0.2 and 0.3 M sorbitol inhibited the growth and maintained the quality of calli and node explants for 12 weeks. It was found that ABA could not inhibit the growth rate after cultures for 12 weeks. Callus and node explant survival from slow growth preservation were able to regrowth which successfully (100%) tested on WPM medium supplemented with 0.25 mg.L-16-benzylaminopurine (BAP) containing 30 g.L-1sucrose after for 2 weeks. - Some of the metrics are blocked by yourconsent settings
Item type:Publication, The effect of plant growth regulator and in vitro conservation of teak (Tectona grandis L.) by tissue culture(2018-12-01) ;Tongsad, P. ;Laipasu, P. ;Chareonsap, P. P.Poeaim, A.The plant regeneration of teak (Tectona grandis L.) was reported. Result showed that the highest number of shoot induction was 55.49 cm., in length of shoots which tested at 0.25 mg·L -1 BAP, and averaged 40.29 cm which tested at 0.25 mg·L -1 mT after 4 weeks. The shoots were elongated using shoot tip explants of cryopreservation on teak by encapsulation-dehydration (EnDe), encapsulation-vitrification (EnVi) and vitrification (Vi) techniques. The shoot tips were used as explants, encapsulated apices were precultured on hormone-free MS medium containing 0.3 M sucrose for 2 days, dehydrated, and immersed in liquid nitrogen for 24 h after that thawed in thermal bath at 40 ºC and washed in unloading solution. Vi and EnVi protocols were used to treat with a loading solution, dehydrated by plant vitrification solution 2 (PVS2). EnDe protocol were encapsulated by desiccated air-dry in laminar air flow and dehydrated with silica gel. Results showed that Vi gave an excellent shoot tip explants for survival and regrowth. Vi were unable to survival. This experiment was preliminary searched to improve and develop the teak for cryopreservation. - Some of the metrics are blocked by yourconsent settings
